Study On The Correlation Between Colorectal Tubular Adenoma With Damp-heat Constitution With The Expression Of Survivin And PTEN

Purpose:To explore the expressions of Survivin and PTEN are correlated with the Colorectal Tubular Adenoma with Damp-heat Constitution?Methods:We randomly selected patients who underwent electronic colonoscopy in the Department of Gastroenterology,Department of Inpatient and Physical Examination of the Second People's Hospital of Fujian Province,and found that colonic polyps were treated with endoscopic polypectomy and postoperative pathological diagnosis of tubular adenomatous polyps.After the judgment of TCM constitution,40 cases were selected as the study group(Damp-heat Constitution),40 cases were included as the control group(Yang-deficiency Constitution),and 20 healthy subjects were selected as the normal control group.The expression levels of Survivin and PTEN in the above three groups were detected by immunohistochemistry.The differences of Survivin and PTEN expression between the three groups were analyzed and compared.Result:1.The age and sex of the Damp-heat constitution,Yang-deficiency constitution and normal control group were compared.The results showed that there was no significant difference in age and gender between the three groups(P>0.05),which was comparable.The distribution of colon adenomas in the study group and the case group was compared.There was no significant difference in the degree of colon adenoma between the two groups(P>0.05),which was also comparable.2.The correlation between Survivin and PTEN expression in all samples was compared with gender,age,and degree of colon adenoma.The results showed that the expression of Survivin and PTEN were not associated with gender and age(P>0.05).However,both of them were related to the degree of adenoma,and the expression of Survivin was up-regulated and the expression of PTEN was decreased with the increase of tumor degree(P<0.05).In the tubular adenoma,the expression of the two was negatively correlated(r_s=-0.665),which was statistically significant(P<0.05).3.The positive expression rate of Survivin was 50.0%(20/40)in Damp-heat constitution and35.0%(14/40)in Yang-deficiency constitution,which was higher than 10.0%(2/20)of normal control group;PTEN positive expression rate was 67.5%(27/40)in Damp-heat constitution and 77.5%(31/40)of the Yang-deficiency constitution were lower than the normal control group of 90.0%(18/20),the difference was statistically significant(P<0.05).In further comparison,the expression of Survivin in the Damp-heat constitution was higher than that in the Yang-deficiency,and the difference was statistically significant(P<0.05).However,the expression of PTEN in the Damp-heat constitution group was lower than that in the Yang-deficiency,but there was no statistical difference(P>0.05).4.Among the colonic tubular adenoma with HGIN,the positive expression rate of Survivin was 78.6%(11/14)in the Damp-heat constitution and 60.0%(6/10)higher than the Yang-deficiency constitution,with statistical difference(P<0.05);Among the colonic tubular adenoma with LGIN,the positive expression rate of Survivin was 34.6%(9/26)in the Damp-heat constitution and 26.7%(8/30)in the Yang-deficiency constitution,no statistical difference(P>0.05).5.Among the colorectal tubular adenoma with HGIN,PTEN positive expression rate in the Damp-heat constitution 50.0%(7/14)is lower than the Yang-deficiency constitution 60.0%(6/10);Among the colorectal tubular adenoma with LGIN,PTEN positive expression 76.9%(20/26)of the rate of Damp-heat constitution was lower than 83.3%(25/30)of Yang-deficiency,but there was no statistical difference(P>0.05).Conclusion:1.In colonic tubular adenomas,especially in HGIN,Survivin expression in Damp-heat constitution is higher than Yang-deficiency constitution,there is a certain correlation,and PTEN expression is not significantly related to Damp-heat constitution.2.In the tubular adenoma of the colon,the expression of Survivin was up-regulated compared with the normal group,and the expression of PTEN was lower than that in the normal group,and there was a significant difference in the degree of tumor differentiation.It is suggested that Survivin and PTEN play a role in the process of adenoma formation from normal mucosa.3?In colonic tubular adenomas,Survivin was negatively correlated with PTEN expression.
