Objectives To investigate the expression of aquaporin 3(AQP3)in placenta and fetal membranes of severe preeclampsia patients and the level of adiponectin in umbilical cord blood,and to analyze the relationship between AQP3,adiponectin and severe preeclampsia.Methods 60 cases severe preeclampsia patients who had delivered in the obstetrics department of Hospital of North China University of science and technology from October 2017 to September 2018 were selected as the case group,and 60 cases of normal pregnant women in the same period were selected as the control group.The clinical data of two groups of pregnant women were collected.The expression of AQP3 in placenta and fetal membranes was determined by immunohistochemistry.The expression level of AQP3 mRNA and protein in placenta and fetal membranes was detected by qRT-PCR and Western blotting,respectively.The expression of adiponectin in umbilical artery blood was detected by ELISA.Independent sample t test was used to compare the measurement data between the case group and the control group,and two independent sample non-parametric test was used to test the grade data,and?~2test was used for counting data.P<0.05 was statistically significant.Results 1 The levels of triglyceride and LDL-C and LDL-C/HDL-C in case group were higher than those in control group(3.89±1.05 vs.2.82±0.98 and 3.81±1.00 vs.3.41±0.85 and 2.06±0.54 vs.1.80±0.52,respectively,P<0.05 for all three of them);Multivariate logistic regression analysis showed that the levels of triglyceride was related to the onset of severe preeclampsia,with OR of 2.589(95%CI 1.348~4.970).2 The results of immunohistochemistry showed that the expression of AQP3 protein in placenta of case group was lower than that of control group(P<0.05);The expression of AQP3protein in fetal membranes of case group was higher than that of control group(P<0.05).3 The results of qRT-PCR showed that the relative expression of AQP3 mRNA in placenta tissue of case group was lower than that of control group(0.81±0.25 vs.1.45±0.18,P<0.05);The relative expression of AQP3 mRNA in the fetal membranes of case group was higher than that of the control group(1.36±0.34 vs.0.77±0.12,P<0.05).4Western blot results showed that the relative expression of AQP3 protein in placenta of case group was significantly lower than that of control group(0.53±0.26 vs.0.83±0.22,P<0.05);The relative expression of AQP3 protein in the fetal membranes of case group was higher than that of the control group(0.97±0.23 vs.0.63±0.21,P<0.05).5 The results of ELISA showed that the level of adiponectin in umbilical cord blood of neonates in case group was higher than that in control group(55.78±3.12 vs.50.31±2.87,P<0.05).Conclusions 1 The expression of AQP3 mRNA and protein was weakened in placenta of severe preeclampsia patients.The expression level of AQP3 protein in placenta is related to the occurrence of severe preeclampsia.When the expression level of AQP3 protein decreases,the risk of severe preeclampsia increases.2 The expression of AQP3 mRNA and protein was increased in fetal membranes of severe preeclampsia patients.3Adiponectin level in cord blood is associated with the onset of severe preeclampsia.Patients with cord blood adiponectin above average had an increased risk of severe pre-eclampsia.Figure 10;Table 15;Reference 104...
