| ObjectiveTo observe the effect of Jiawei Zengye Decoction on the expression of AQP4 and VIP in colonic tissue of rats with slow transit constipation at molecular level,and to analyze the regulatory mechanism of AQP4 and VIP in improving STC symptoms,so as to provide some reference for STC related pathway research.Method1.STC rat model was established by compound diphenoxylate,and the effect of STC rat model was evaluated by statistical method;2.After drug intervention,the colon histopathological changes of STC rats were observed by hematoxylin-eosin staining;3.Immunohistochemistry and Western blot were used to detect the expression of AQP4 and VIP in colon of STC rats after drug intervention.Result1.STC rat model was established by compound diphenoxylate.The body weight of STC rats increased slowly compared with the normal group(P < 0.001),and the amount of feces and water content of feces decreased significantly in 24 hours(P < 0.01);2.After drug intervention,hematoxylin-eosin staining showed that the colonic structure of rats in the normal group was intact,the colonic mucosa of rats in the model group was ruptured and exuded,while the other groups had less exudation;3.Compared with the model group,the contents of AQP4 and VIP in colon tissue of STC rats decreased significantly after the intervention of Jiawei Zengye Decoction(P < 0.05).Conclusion1.Jiawei Zengye Decoction can alleviate the constipation symptoms of STC rats,increase the body weight of STC rats,gradually slow down the decreasing trend of feces in STC rats,increase the water content of feces and improve the intestinal transmission function;2.Jiawei Zengye Decoction can down-regulate the expression of AQP4 and VIP in colon tissue of STC rats,regulate intestinal water metabolism,and increase the water content of rat feces;3.The consistency of AQP4 and VIP in the experiment improves the STC symptoms.We can use AQPs-VIP as a pathway for STC experimental study. |
