| Objective:To observe the effect of Ancient Han Healtn Essence on the expression of IL-6,TNF-?and HMGB1 in asthma with kidney-yang deficiency.Methods:30 healthy male wistar rats aged 4 weeks,weight of?120±10?g,were randomly divided into 6 groups?n=5 per group?:normal group?A group?,asthma group?B group?,asthma with kidney-yang deficiency group?C group?,Ancient Han Healtn Essence low,medium and high does+asthma with kidney-yang deficiency group?D,E,F group?Establishment of kidney-yang deficiency model by intramuscular injection of hydrocortisone injection 25mg.kg-1.d-1 in C-F group.The B-F group was sensitized by intraperitoneal injection of a mixture of OVA1mg+Al?OH?100mg saline to 0.1ml mixture for many times to establish asthma model.A group was sensitized by intraperitoneal injection of normal saline 0.1ml,and distilled water was inhaled as control.In D-F group,the corresponding dose of Ancient Han Healtn Essence was given to each group half an hour before each atomization stimulation?4 ml/kg,8ml/kg and 16 ml/kg,respectively?for 14 consecutive days.Group A-C was given the same amount of saline.24 hours after the last atomization,rats were killed by anesthesia and their lung tissues were taken out.The pathological changes of lung tissues were observed by HE staining,the expressions of IL-6 and TNF-?in lung tissues were detected by immunohistochemistry,and the relative expressions of IL-6,TNF-?and HMGB1 in lung tissues were detected by QPCR.Results:1.HE staining showed that the structure of bronchus was normal and the layers were clear,there was no obvious infiltration of inflammatory cells,and the airway mucosa was intact in the normal group.In asthma group,infiltration of inflammatory cells,irregular lumen,edema and thickening of bronchial wall and interruption of mucosal fold were observed in bronchial tissue.The lung tissue of rats in the asthma group with kidney-yang deficiency showed more bronchial folds,narrower lumen,the infiltration of peripheral inflammatory cells was higher than that of asthma group,and the thickness of airway wall was more obvious than that of asthma group.The infiltration of inflammatory cells and mucosal fold in Ancient Han Healtn Essence group were lower than those in kidney-yang deficiency asthma group,the lumen was still narrower than that in normal group,and the mucosal fold was slightly interrupted.The severity of the lesion was lighter than that of the asthma group with kidney-yang deficiency,but there was no significant difference between the different concentration groups.2.Immunohistochy showed that IL-6 and TNF-?proteinthe expression of IL-6 and TNF-?protein in asthma group with kidney-yang deficiency and asthma group was higher than that in normal group,and there was a significant difference between the two groups?P<0 01?.and the expression of IL-6 and TNF-?protein in asthma group with kidney-yang deficiency was more significant than that in asthma group?P<0.05?.The expression of IL-6 and TNF-?in the low,medium and high dose groups of Ancient Han Healtn Essence was significantly lower than that in the asthma group with kidney-yang deficiency?P<0.01?,but there was no significant difference among the low,medium and high dose groups of Ancient Han Healtn Essence group?P>0.05?.3.QPCR showed that the relative expression of IL-6,TNF-?and HMGB1 increased in asthma group and asthma group with kidney-yang deficiency,which was significantly different from that in normal group?P<0.05?.The expression of HMGB1,IL-6 and TNF-?in asthma group with kidney-yang deficiency was higher than that in asthma group?P<0.05?.The expression of IL-6,TNF-?and HMGB1 mRNA in the low,middle and high dose groups of Ancient Han Healtn Essence were lower than those in the Asthma group with kidney-yang deficiency?P<0 01?,but there was no significant difference among the three groups?P>0 05?.Conclusions:1.The expression of IL-6,TNF-?and HMGB1 are increased in asthmatic rats with kidney-yang deficiency.2.The Ancient Han Healtn Essence can improve the inflammatory response of asthmatic rats with kidney-yang deficiency,the mechanism may be related to inhibiting the expression of IL-6,TNF-?and HMGB1. |
