Objective:We aim to detect the expression of miRNA-132-5p and miRNA-133a-3p in patients with coronary heart disease,so as to provide experimental basis for clinical discovery of new biomarkers.Methods:We collected 72 patients with chest tightness or chest pain as the chief complaint in the affiliated cardiovascular hospital of xiamen university from September 2017 to October 2018,including 24 cases of acute myocardial infarction,23 cases of unstable angina and 25 cases of non-coronary heart disease.We collected general data and clinical test indicators,and measured the expression of miRNA by real-time fluorescence quantitative pCR.Results:1.WBC count,glutamic-pyruvic transaminase,urea nitrogen,creatinine,troponin in AMI(Acute myocardial infarction)group were higher than that in Non-CHD(Non-Chronic coronary artery disease)group(p<0.05),WBC count,glutamic-pyruvic transaminase,urea nitrogen,creatinine,troponin in AMI group is higher than that in the UA(Unstable angina pectoris)group(p<0.05).WBC count,NT-proBNP in UA group of was higher than that in Non-CHD group(p<0.05).2.The relative expression of miRNA-132-5p in AMI group and UA group was lower than that in Non-CHD group(p<0.05),and that in AMI group was lower than that in UA group(p<0.05).The relative expression of mRNA-133a-3p in the AMI group was higher than that in the Non-CHD group(p<0.0001),and that in the AMI group was higher than that in the UA group(p<0.05).3.We compared AMI group with Non-AMI group(UA group+Non-CHD group),ROC curve analysis showed that the AUC of miRNA-132-5p was 0.8793(95%CI:0.7977-0.9610,p<0.0001),with a sensitivity of 83.33%and a specificity of 87.50%.The AUC of miRNA-133a-3p was 0.8949(95%CI:0.8140-0.9759,p<0.0001),with a sensitivity of 87.50%and a specificity of 78.26%.We combined miRNA-132-5p and miRNA-133a-3p to diagnose AMI,the AUC was 0.9540(95%ci:0.9093-0.9987,p<0.0001),and the diagnostic sensitivity and specificity were 91.67%and 91.67%,respectively.4.The relative expression of miRNA-132-5p was negatively correlated with WBC count and blood creatinine,while the relative expression of miRNA-133a-3p was positively correlated with WBC count,urea nitrogen and blood creatinine.Conclusion:1.Compared with Non-CHD group,the expression of miRNA-132-5p increase,and that of miRNA-133a-3p decrease in the AMI group and UA group.2.MiRNA-132-5p and miRNA-133a-3p are potential biomarkers for the diagnosis of acute myocardial infarction,we can combine them to improve sensitivity and specificity for diagnosis.
