Objective:To investigate hsa-miRNA-133a expression characteristics and its bioinformatics of esophageal squamous cell carcinoma(ESCC) in Huaian China.Methods:The expression patterns of hsa-miRNA-133a in patients with ESCC in Huaian were assessed through Microarray Assay and qRT-PCR. The sequence of hsa-miRNA-133a was got from miRBase database, NCBI, UCSC Browser, and targeted genes of hsa-miRNA-133a were predicted by miRanda, TargetScan, DIANA LAB-TarBase5.0 and miRTarBase, and then analyzed by GO (gene ontology) and Pathway analysis. Dual-lueiferase reporter system was used to identitify the direct target genes of hsa-miRNA-133a. The level of VEGFA after overexpression/downregulation of hsa-miRNA-133a in human ESCC cells were detected by Western BlotResults:1. Compared with the paired normal tumor-adjacent tissue, the expression of hsa-miRNA-133a in ESCC was significantly downregulated by Microarray Assay and qRT-PCR(P<0.01) in Huaian; 2. The expression of hsa-miRNA-133a in lymph node metastasis group were significantly lower than negative group (P<0.05).3. Compared with normal esophageal epithelial cell lines Het-1A, the expression levels of hsa-miR-133a were significantly lower in human ESCC cell lines KYSE-150, ECA109; 5. Hsa-miRNA-133a was highly conserved among species. Gene ontology analysis demonstrated that the targeted genes of hsa-miRNA-133a were enriched in protein modification, cell proliferation and apoptosis, and other biological processes (P<0.001). The pathway analysis indicated that the targeted genes were mainly involved in cancer signaling, apoptosis signaling, and other pathways (P<0.05).6. VEGFA involved in tumor metastasis could be a target gene of hsa-miRNA-133a.Conclusion:1. The expression of hsa-miRNA-133a in ESCC was significantly lower than that in the paired tumor-adjacent tissue.2. The expression of hsa-miRNA-133a in lymph node metastasis group were significantly lower than negative group, which illustrated that downregulation of miRNA-133a might be related to the migration of human ESCC.3. VEGFA might be involved in hsa-miRNA-133a mediated ESCC metastasis.
