Recurrent spontaneous abortion(RSA) means contimuous 3 times or more than 3 times repeatedly happened early miscarriage except all known reasons, and it is one of common diseases of pathological pregnacy. Pathogeny of RSA is very complex, including chromosomal abnormalities, infection factors, endocrine abnormalities, immune factors, and uterine anatomic factors, etc. However, the etiology in 50% patients is still unknown. In recent years, the relationship between the micro RNAs(mi RNA) and human diseases gradually gets more and more attention, and its important role in pathogenetic development is proving step by step. More and more studies have proved that mi RNA played an important role in tumors, congenital heart disease, blood diseases, neurological diseases and other diseases. Nowadays, the abnormal expressions of mi RNAs in RSA have gradually attracted the attention of scholars.Objective: To observe and compare the changes of mi RNA-133 a, mi RNA-135 a and mi RNA-135 b contents in chorionic villi tissues of RSA group and normal early pregnancy group, and to investigate the different expressions in the chorionic villi tissues of these two groups, so as to provide the theoretical and experimental basis for RSA pathogenesis.Methods: A total of 17 RSA patients were recruited as research group and 18 normal early pregnancy women as control group. In situ hybridization and the real-time PCR methods were used to investigate the expressions of mi RNA-133 a, mi RNA-135 a and mi RNA-135 b in the chorionic villi tissues of RSA group and normal early pregnancy group.Results:1 In situ hybridization showed that the mi RNA-133 a, mi RNA-135 a and mi RNA-135 b were all expressed in the two groups, and expressions of mi RNA-133 a and mi RNA-135 a in the chorionic villi tissues of RSA group were significantly higher than those in normal early pregnancy group(P<0.05), however, there was no statistically significant difference between RSA group and normal early pregnancy group in expression of mi RNA-135b(P>0.05).2 Real-time PCR showed that the mi RNA-133 a, mi RNA-135 a and mi RNA-135 b were all expressed in the two groups, and expressions of mi RNA-133 a and mi RNA-135 a in the chorionic villi tissues of RSA group were significantly higher than those in normal early pregnancy group(P<0.05), however, there was no statistically significant difference between RSA group and normal early pregnancy group in expression of mi RNA-135b(P>0.05).Conclusions:1 The mi RNA-135 b is expressed in the chorionic villi tissues of both RSA and normal early pregnancy.2 The increased expressions of mi RNA-133 a and mi RNA-135 a in the chorionic villi tissues of RSA suggested that the mi RNA-133 a and mi RNA-135 a might take part in the occurrence of RSA.
