The chemical characteristic fingerprint of The traditional Chinese medicine(TCM)method has become one of the most effective way internationally recognized quality control of Chinese herbal medicine and natural medicine. Hydrogen nuclear magnetic resonance(1H-NMR) technology can theoretically detect any hydrogen atom contained in organic components. The paper selected the Eucommia ulmoides as the research object, By means of chemical plants get the monomer compounds. Through the chemical shift, preliminary belonging the characteristic compounds, and through the methodology study, further verify the correctness of the characteristic peak of belonging.The dried and crushed bark of Eucommia ulmoides was extracted with 95% alcohol by reflux, the solvent was concentrated to extractum by reduced pressure, then extracted with different solvents. A variety of chromatographic method was used to separate compounds, and the chemical structures were identified by spectroscopic techniquesand and physical and chemical properties. 16 compounds were obtained from the bark of Eucommia ulmoides Oilv, they were identified as β-sitosterol(1),cycloeucalenol(2), betulinic acid(3), 24-methylenecycloartenone(4),cycloeucalenone(5), salicifoliol(6), pinoresinol(7), genipin(8), alternariol(9),balanophonin(10), eucommidiol(11), pinoresinol-4’-O-β-D-glucopyranoside(12),eucommiol(13), deoxyeucommiol(14), 8-hydroxypinoresinol(15) and dehydrodiconiferyl alcohol γ’-O-β-D-glucopyranoside(16), compounds 2, 4, 5, 6, 9,10, 16, was isolated from this genus for the first time, compounds 14 was isolated from the bark of plant for the first time. Viaing the studying of extracting solvent,solvent polarity, extraction conditions, and the use of CPMG technology for simplifying the map, preliminary belonging PDG and eucommidiol, In order to make the characteristic peak of identify more reliable, using titration, STOCSY,two-dimensional HMQC and HMBC spectrum and the two-dimensional J-coupling spectrum, with pinoresinol-di-O-β-D-glucopyranoside and eucommia as the standard,study the methodology on belonging characteristic peaks. Results show that thecharacteristic peak of belonging is accurate and consistent, this also confirmed the series of method is feasible and scientific.
