In the dissertation,Eucommia ulmoides in different harvesting years and different medicinal parts is used as the main research part.The content of the main active ingredients in different compounds:lignans,phenylpropanoids,iridoids and flavonoids is determined.Analyze the spatial and temporal distribution of these active ingredients.Correlation analysis was then used to study the differences in vasodilation effect of Eucommia ulmoides in different harvesting years and different medicinal parts.The main research contents and results are as follows:1.The HPLC method was used to change the wavelength and perform gradient elution,and then the 12 active ingredients in iridoids,lignans and phenylpropanoids of Eucommia ulmoides were measured simultaneously.Simultaneous determination of five active ingredients in flavonoids in Eucommia ulmoides.The experimental results show that the RSD(%)of the precision,repeatability,stability,and recovery of the 17 active ingredients of Eucommia ulmoides by two HPLC methods are all within 5%.We performed linear regression on standards of different concentrations and found that their correlation coefficients were all above 0.999.Correlation analysis was performed on the 17 main active ingredients with harvesting years and medicinal parts through heat maps,and the results showed that the active ingredients in iridoids,lignans,and phenylpropanoids will vary depending on Differences in harvesting years and locations.2.We have studied the active ingredients in lignans,iridoids,phenylpropanoids and flavonoids in Eucommia ulmoides with different medicinal parts and different harvesting years.The results show that the content of these active ingredients in Eucommia ulmoides will vary with the harvest time of Eucommia ulmoides.The average content of pinoresinol diglucoside in the Eucommia ulmoides crust from 20 to 25 years is higher than that of other parts and have significant difference(P<0.01).The content of Aucubin in the Eucommia ulmoides skin after 21 years is lower than that in other parts and have significant difference(P<0.01).The content of chlorogenic acid in Eucommia ulmoides leaf at 16 and 24 years is higher than the other two parts and have significant difference(P<0.01).The content of Hyperoside in Eucommia ulmoides leaf from 16 to 25 years is higher than the other two parts and have significant difference(P<0.01).Eucommia ulmoides at different harvesting locations will affect the content of active ingredients in lignans,ridoids,phenylpropanoids and flavonoids.There are distribution rules between them and the rules are different.The average content of pinoresinol diglucoside is Eucommia ulmoides crust>Eucommia ulmoides skin>Eucommia ulmoides leaf.The average content of Aucubin is Eucommia ulmoides crust>Eucommia ulmoides leaf>Eucommia ulmoides skin.The average content of Chlorogenic acid is Eucommia ulmoides leaf>Eucommia ulmoides crust>Eucommia ulmoides skin.The average content of Hyperoside is Eucommia ulmoides leaf>Eucommia ulmoides skin>Eucommia ulmoides crust.3.We explored the differences in vasodilation of Eucommia ulmoides in different harvest years and medicinal parts.The results showed that Eucommia ulmoides samples had no contractile effect on the normal tension of the untreated vascular ring.Eucommia ulmoides samples had a significant diastolic effect on KCl-induced contraction of isolated thoracic aorta vascular rings in rats,and its diastolic effect is concentration-dependent.Different Eucommia samples had different abilities to activate the VOCC channels of the cell membrane,and the diastolic effect might be related to the decrease in the calcium concentration in the vascular ring.Different Eucommia ulmoides samples had different abilities to inhibit the release of vasodilation factors NO and PGI2,and thus produced different degrees of diastolic effect.4.Taking the active ingredients in the four types of active compounds in Eucommia ulmoides in different harvesting years and different medicinal parts as research objects,the principal component analysis and correlation analysis were performed on them respectively.The results showed that among the lignans of Eucommia ulmoides in different harvest years and medicinal parts,Syringaresinol Diglucoside(SDG)in the KCL and Ca.groups had better correlation with vasodilation.Pinoresinol Diglucoside(PDG)in the Indo group had better correlation with vasodilation.The samples with better antihypertensive effects are:18-b,21-c,22-b,23-c,24-c,25-c.Among the iridoids of Eucommia ulmoides in different harvesting years and medicinal parts,Aucubin and Geniposidic Acid in the L-NAME group had lower correlation with vasodilation.The samples with weaker antihypertensive effects are:18-b,25-a,16-b,17-c,18-c,19-c.Among the phenylpropanoids of Eucommia ulmoides in different harvesting years and medicinal parts,Caffeic Acid and Procatechin Acid in the Indo group had better correlation with vasodilation.The samples with better antihypertensive effects are:23-c,24-c,20-c,23-b.Among the flavonoids of Eucommia ulmoides in different harvesting years and medicinal parts,Quercitrin and Rutin in the L-NAME group had better correlation with vasodilation.The samples with better antihypertensive effects are:23-b,23-c,24-c,17-c.
