| Eucommia ulmoides Oliver is a tertiary relic plant that only exists in China,and it’s bark is a traditional and precious Chinese medicine.Eucommia ulmoides mainly contains organic compounds such as lignans,iridoids,phenolics,flavonoids,steroids,terpenoids and inorganic trace elements such as calcium and iron.Eucommia ulmoides seeds,known as Samaras in the botanical category,are rich in a variety of natural active ingredients such as oil,protein,iridoids,and the like.In this paper,the chemical constituents in defatted Eucommia ulmoides seed meal were extracted and separated,and the structure was identified.The compounds with large amount and good activity were modified to obtain better compounds.The main findings are as follows:(1)Separation and purification of n-butanol phase compounds in defatted Eucommia ulmoides seeds by a variety of modern chromatographic separation technologies(positive/reversed-phase silica gel,Sephadex LH-20,LC-MS,HPLC),and use a variety of spectroscopy methods(NMR,IR,UV,CD,HRMS)for structural identification of the isolated compounds.A total of 7 iridoids were obtained:Aucubin(1),Bartsioside(2),Linaride(3),Geniposidic acid(4),Scyphiphin D(5),Ulmoidoside A(6),Ulmoidoside B(7),of which the compounds Linarid(3)and Scyphiphin D(5)were isolated from Eucommia for the first time;(2)The obtained compounds are tested for biological activity,including neurotrophic activity,anti-inflammatory activity andα-glucosidase inhibitory activity.the anti-inflammat ory activity results show that:using BV-2 cells as a model,the tested compounds have obvious anti-inflammatory activity at a concentration of 10μM,the inhibition rate of nitric-oxide production is 66%-75%,Compound 1 has the highest anti-inflammatory activity with IC50 value of 29.25μM.The tested compounds did not have good neurotrophic activity andα-glucosidase inhibitory activity;(3)The structural modification of aucubin(1)and baltiin(2)resulted in a total of 8derivatives including 6 new compounds.modeled on microglia(BV-2)at a concentration of10μM,The results of the activity test showed that none of the semi-synthetic compounds had anti-inflammatory activity;modeled on macrophages at a concentration of 10μM,The results of the activity test showed that the semi-synthetic compound has only weak anti-inflammatory activity. |
