Background: Post-stroke anxiety disorder(PSAD)is a high incidence,clinical workers are easy to be neglected affective disorder after stroke,can affect the functional recovery of patients,prolong the hospital stay,even can cause of recurrence of stroke.AS the manifestations of PSAD is not typical and almost of them are mild case,easy to be ignored,without safe and effective treatment,therefore understanding the pathogenesis of anxiety disorder after stroke to early identify patients at high risk of anxiety disorders in stroke patients and provide new therapeutic targets for the treatment,has important practical significance.MiRNA is involved in the regulation of human genes and is closely related to the occurrence and development of diseases.The purpose of this study was to understand the specific miRNA expression profile of post-stroke anxiety disorder,and its role in the pathogenesis of post-stroke anxiety disorder.At present,there are no studies on miRNA of post-stroke anxiety disorder.Method:We include hospitalized patients in GuangXi medical university first affiliate neurology from September,2017 to March 2018,divided into after stroke without anxiety group(hereinafter referred to as stroke group),post stroke anxiety group and normal control group in the light of clinical assessment.Extracting micrornas from peripheral blood and sequencing,screening differentially expressed micrornas,verifying its expression level by real-time fluorescent quantitative polymerase chain reaction(QRT-PCR).The function of miRNA with differential expression was analyzed by bioinformatics analysis.Result: Through high-throughput sequencing,an average of 12672,469 Raw Data was measured in each sample,and 11981,356 clean Data were obtained from each sample after screening.Results suggest the stroke group and normal control group with significant differences 68 miRNA expression,the post-stroke anxiety group and normal control group with significant difference exists among 47 miRNAs expression,post-stroke anxiety group and stroke group 37 miRNAs expression exists significant difference between,there are two miRNAs expression differences in each group,which is mir-206 and mir-144-5p respectively.Further PCR quantitative validation of the differentially expressed mir-206 and mir-144-5p.There were significant differences in the expression of mir-206 in peripheral blood between the three groups(P= 0.014,P <0.05).Compared with the normal control group,the expression of mir-206 in peripheral blood was down-regulated in the stroke group(P=0.007),while the expression of mir-206 in peripheral blood of the post-stroke anxiety group was down-regulated,but there was no statistical significance(P=0.587).Compared with the stroke group,the expression of mir-206 in peripheral blood of the post-stroke anxiety group was up-regulated(P=0.022),while the expression level of miR-144-5p between groups show no difference.According to the bioinformatics prediction mir-206 was involved in the regulation of 30 signaling pathways,of which the neurotrophic signaling pathway may be associated with the occurrence of post-stroke anxiety disorder.Conclusion: There was a specific miRNA expression profile in post-stroke anxiety disorder.The expression of mir-206 in the blood of patients with post-stroke anxiety disorder was increased,and the expression of BDNF could be reduced to participate in the onset of anxiety disorder after stroke.
