Cardiovascular disease is a common and frequently-occurring disease,its morbidity and mortality have remained high.RNA-binding proteins play an important role in the regulation of post-transcriptional gene expression by binding RNA.Many RNA binding proteins have been reported to be closely related to cardiac development and cardiovascular disease.We preliminarily investigated the function of two RNA-binding proteins,RBM24 and RBM47,in heart disease.RBM24 is a key regulator of cardiac development and sarcomere assembly,and it is also recognized by the American Heart Association as a potential marker and one of the major risk factors for coronary heart disease.Now,we found that the serine at 181 of RBM24 can be phosphorylated by GSK3?,and this phosphorylation is significantly activated in the heart of stressed mice.We hypothesized that the regulation of RBM24 phosphorylation could mediate the effect of mental stress on heart diseases.RBM47 is able to inhibit the Wnt signaling pathway and inhibit tumor progression by modulating alternative splicing of multiple RNAs.And RBM47 plays a crucial role in the development of the zebrafish's head.In our study,we found that the expression level of RBM47 was significantly up-regulated in the model of cardiac injury regeneration both in zebrafish and newborn mice.We speculate that RBM47 plays a role in cardiac regeneration.Overall,this thesis will study the role of RNA-binding proteins(RBM24 and RBM47)in heart disease.
