objective:To observe the concentration of B-Type natriuretic peptide (BNP) and heart-type fatty acid-binding protein (HFABP) in different heart function, to explore the meaning in the chronic heart failure and the relationship to traditional Chinese medicine-heart yang failure of the rats.Methods:40 convetional SPF male wistar rats,weight(200±20)g,were randomly divided into chronic heart failure (CHF) model group,30 rats;physiologic scaline (NS) (normal control group),10 rats.1.CHF model established:from 30 healthy male rats,weight(200±20)g,inject pentobarbitat(20-25mg/kg) in the abdomen, then inject isoprotereno (ISO)(Preparation of 85mg/kg weight,repeated after 24 hours)into hypoderm every one; and devided the survival into three groups:CHF 2 weeks,4 weeks and 8 weeks.2.NS group to normal:from 10 healthy male rats,weight(200±20)g,injected NS 0.25 ml into hypoderm every one, repeat after 24 hours.3.Model identification:identification to the models in two weeks, four weeks and eight weeks.(1)cardiac function testing:by carotid artery,measurement of left venricular systolic pressure (LVSP),left ventricular end-diastolic pressure (LVEDP) and the largest increase,rise rate or decline of left ventricular pressure(±LVdp/dt).(2) determination of plasma HFABP and BNP by enzyme-linked immunosorbent assay (ELISA).(4)Biopsy pathologytest:Left myocardial tissue:conventional paraffin-embedded sections,HE staining,microscopy.4.Left ventricular mass index(LVMI) determination,liver and lung mass index.5.Data analysis:Group t-test, Correlation analysis using linear correlation analysis, p<0.05 for the difference w-assignificant. statistical analysia by SPSS 13.00.Result: 1.general state of health, death rate:CHF model group, two rats died every group, the mortal rate was 20%,the cause of death was considered of congestive heart failure,or arrhythmia, and the NS group with no death. And the rate of groups was p<0.05;CHF rats:the spirit was weaker,and ate less than the NS group,they are less active.the urine decreased.the fur was lack of brightness.the lip was cyanosis, their weight got less increased. The symtoms showd the rats had been heart yang failure.2. Heart changes of pathologic anatomy:the specimen of the heart in the sight:the surface of the NS group seemed smooth,the colour was red while CHF group seemed gloomy and the colour was violetly deep. morphology pathological changes of rat myocardial tissue,results of HEstaining:HEstaining showed, CHF group:(1)myocardial tissue was damaged:some of the myocardial fiber arranged out of order and get corpulent,the cell was atrophy and degeration; (2) some of the myocardial fiber was dissolved or broken,some extracellular matrix was found bloody and fibrosis;Connective Tissue and fibroblast was proliferate. (3)Leucocyte was nfiltration in. rat myocardial fibers were neat and the nucleus were clear,cleatrance of normal cells,no myocardial fiber damaged; 3.Determination of hemodynamics: compared with the NS group,there was no difference between NS group and CHF model two weeks;other CHF groups' LVSP,±LVdp/dtmax was significantly decreased (p<0.05), LVEDP was significantly increased (p<0.05);4.Ventricular mass index of four groups rats:compared with the NS groups, there was no difference between NS group and CHF model two weeks;other CHF groups increased(p<0.01); among the CHF groups, the index increased (p<0.01).4.Plasma heart-type fatty acid-binding protein in rats, compared with the NS group,CHF groups HFABP increased, CHF groups also increased significantly by the time.and the difference is significantly (p<0.01);5. Plasma B-Type natriuretic peptide(BNP) in rats,compared with the NS group,CHF groups BNP were increased, CHF groups also increased significantly by the time.and the difference in both NS group and CHF group(two weeks) was significantly (p<0.05);also was in both CHF groups(two weeks and four weeks);and among other groups,their diference was significantly(p<0.01).Conclusions:1.from the normal group to the heart failure group, Plasma B-Type natriuretic peptide concentration increased, Plasma B-Type natriuretic peptide concentration of the rats in corresponding with cardiac homogenates were positively correlated. r=-0.855.2.from the normal group to the heart failure group, plasma heart-type fatty acid-binding protein concentration of the rats in corresponding with cardiac homogenates were positively correlated. r=-0.893.3.plasma heart-type fatty acid-binding protein concentration of the rats in corresponding with plasma B-Type natriuretic peptide were positively correlated, r=0.914.4.from CHF model,we could find cardiac fibrosis got more and more obvious.and myocardial hypertrophy changed there,the extent of CHF got more and more servious.
