objective: 1.Combined with modern data mining methods,discuss the medication law of Professor Wu Shengyuan in the treatment of osteoarthritis,inherits the academic thought and clinical experience of famous old traditional Chinese medicine,provides a reference for the clinical treatment of osteoarthritis.2.To explore the regulatory effect of Yiqi Yangxue Decoction on Wnt/?-catenin signaling pathway,and to provide theoretical basis for Yiqi Yangxue Decoction in the treatment of osteoarthritis.Methods:1.A total of 395 cases of osteoarthritis were elect,which is from My record from March 2016 to September 2016 and the osteoarthritis patient treated in Professor Wu Shengyuan outpatient Department of Yunnan Provincial Hospital of traditional Chinese Medicine from 1997 to 2016.Input the software of traditional Chinese medicine inheritance assistance system(v2.5),carry on the data mining to the screened medical record,analyze its prescription rule.2.Routine preparation of drug serum.The chondrocyte system of knee joint cartilage of 40 SD rats was established in vitro.the chondrocytes were randomly divided into normal group and induced group.HE staining,Alicolan staining and immunofluorescence were used to identify the morphology of chondrocytes and draw the growth curve of chondrocytes.Chondrocytes induced by IL-1 ? were divided into 8 groups: Yiqi Yangxue Decoction drug serum group(20%,10%,5%),saline serum group(20%,10%,5%),Positive control group(drug DKK-1 30ng/ml)and IL-1 ?(10ng/ml)induced group,Apoptosis positive control group(treated with DNA enzyme)and cell culture medium control group were set up.The effect of drug-containing serum on chondrocyte proliferation was observed.RT-qPCR and western blot methods were used to clarify the regulatory effect of Yiqi Yangxue Decoction on the expression of?-catenin,WISP-1,MMP-1 and MMP-13 in rat osteoarthritis model induced by IL-1? from the level of mRNA gene and protein.Results:1.Based on the analysis of 395 prescriptions,46 kinds of drugs were obtained for the treatment of osteoarthritis more than 10 times,among which the drugs belonging to spleen,lung,liver and kidney were the most.Warm medicine and sweet medicine are the most widely used.The core combination of 33 groups was obtained,and a new prescription for the treatment of osteoarthritis was obtained.2.Compared with the normal group,the cartilage morphology of the induced group changed significantly,the cell growth was slow.Compared with IL-1 ? induced group,cartilage proliferation was significantly different in 10% and 20% Yiqi Yangxue Decoction drug-containing serum groups(P < 0.05,P < 0.05),and the expression of ?-catenin,WISP-1,MMP-1,MMP-13 mRNA and protein was down-regulated(P < 0.05,P < 0.05).Conclusion:1."Regulating and tonifying Zhongjiao" and "Use the drug of Gan and Xin" are the characteristics of Professor Wu Shengyuan's differentiation and treatment of osteoarthritis.Focus the "making good use of classical prescriptions and using drugs live",forms a unique diagnosis and treatment experience for Professor Wu Shengyuan.2.Yiqi Yangxue Decoction may regulate Wnt/?-catenin signaling pathway by down-regulating ?-catenin and WISP-1,and inhibit downstream MMP-1and MMP-13 levels,promoting chondrocyte proliferation,which may be one of the mechanisms of Yiqi Yangxue Decoction in reducing articular cartilage destruction.
