Objective: To explore the effect of Yi Qi Yang Xue Decoction on cartilage degeneration of knee osteoarthritis(qi deficiency syndrome)from the point of view of the balance regulation of Wnt/beta-catenin signaling pathways regulating.Then revealing itstreatment of the partial mechanism of osteoarthritis(OA),and providing a theoretical basis for the Yi Qi Yang Xue Decoction in treating OA.Methods: 1 Through an experiment using Sodium iodoacetate,papain,type ii collagen enzyme,surgery Hulth KOA model is set up,HE staining,observe the pathological,in order to select the build mould method.2 110 rats were randomly divided into Yiqi Yangxue Decoction of low,middle,high dosage groups,the model group,positive control group,sham operation group and normal group,Each group have 15 rats.The model of Hulth method were replicated in rats except the normal group and sham operation group.The sham operation group only cut the joint capsule,and not to cut off ligament and meniscus.They took Yiqi Yangxue Decoction(low 1.89g/kg.d,middle 3.78g/kg.d,high 7.56g/kg.d),Didofence Sodium Enteric-coated Tablets(4mg/kg.d),physiological saline(2.5ml.d).The rats normal were given conventional feeding.After drug intervention by 8 weeks,Take the right knee,HE staining was adopted.By gross observation,Pathological scale was used for measuring the cellular morphology of articular cartilage.Through real-time fluorescent quantitative PCR to calculate the level of ?-catenin?MMP-13?BMP-4and BMP-2m RNA.ELISA was used to detect the levels in serumof Wnt3?Wnt4?Wnt5?DKK1?s FRP3 and WIF1;Results: 1.Compared with normal group,the Hulth model of surface crack defects of articular cartilage,disordered arrangement of cells;The rest of the group of articular cartilage surface smooth,neat cells.2.Compared with normal group,sham operation group,model group rats crack defect of articular cartilage,disordered arrangement,the model of Hulth is successful.The joint pathological score is higher(P <0.05).The level of beta-catenin,BMP-2,BMP-4 m RNA in articular cartilage is in significantly increased.The level of Wnt4 in serum of rats in model group is rising,and the numerical in WIF1 and SFRP3 is lower(P < 0.05);Compared with model group,Yiqi Yangxue decoction can reduce the KOA pathological grading of articular cartilage in rats.At the same time it can reduce the levels of beta-catenin,BMP-2,BMP – 4m RNA(P < 0.05),especially in high dose group is more obviously(P < 0.01).Yiqi Yangxue decoction in the middle,high dose group could reduce the content of Wnt4,increase the content of WIF1?SFRP3.Compared with positive control group,the high dose fo Yiqi Yangxue decoction has a better effect on reduing the level of beta-catenin,BMP– 2m RNA.The MMP-13 has no statistical significance.Conclusion: 1.Hulth method conforms to this topic after the experiment model selection.2.Yiqi yangxue decoction could decline the pathological grading score of rats,promote osteoarthritis degenerative cartilage;Yiqi Yangxue decoction could rise WIF1 and s FRP-3,reduce the expression of Wnt4,inhibit the expression of beta – catenin? BMP – 2?BMP-4m RNA.So as to to regulate the balance of Wnt/beta-catenin signaling pathway,Thus inhibiting cartilage matrix degradation,delay cartilage degenerative osteoarthritis,it may be one of the mechanisms of Yiqi Yangxue decoction for the treatment of OA.
