Jianpi Shengxue Granules are a Chinese Medicine and Western Medicine c ombined drug which consists of Ferrous sulfate embedding by?-cyclodextrin a nd Vitamin C on the basis of Shenling Baizhu powder?consists of many TCM such as Yams,Radix Ophiopogonis,Poria cocos etal.?,this preparation has obvi ous effect not only on treating kids'iron-deficiency anemia caused by heart-splee n deficiency and spleen-stomach deficiency,but also on adults'iron-deficiency anemia caused by deficiency of qi and blood.The preparation is a chemically complex exforge,full of flavone,organic acid,saponin and many others chemical constituents,this paper conducts a systematic research on the chemical constituents of Jianpi Shengxue Granules in oder to il luminate its substance foundation.This paper is a foundation for the study of thes cientific implication about the compatibility of medicines and the modernization of TCM.The extractum of Jianpi Shengxue Granules were extracted by the organic solvent extraction methods with three different solvents?PE,EA and butanol?and gots three different polarity parts.The chemical constituents of ethyl acetat e extract and butanol extract were studied but no petroleum ether extract on ac count of its less quality?2g?.The macroporous resin,silica gel column chromatography and semi-prepart ive high performance liquid chromatography were used to isolate and purify th e chemical constituents of Jianpi Shengxue Granules on the basis of the overvi ew of the chemical constituents and pharmacological action of the main single herb in the prescription of Jianpi Shengxue Granules.19 compounds are obtain ed in total.Then all the 19 compounds were identified with ¹H-NMR,¹³C-NMR,MS and their physical-chemical properties.They are palmitic acid?1?,adenine?2?,catechin?3?,glycyrrhizin?4?,isoquiritigenin?5?,formononetin?6?,3?-meth oxydaidzein?7?,oleanolic acid?8?,Schisantherin A?9?,schisandrin B?10?,dio sgenin?11?,succinicacid?12?,5-hyoxymethyl-2-furfura?13?,uridine?14?,glycyrrh etinic acid?15?,3',4'-dihydroxy-7-methoxyflovonol3-O-?-D-glucoside?16?,4'-hydr oxy-flavanone-3-O-?-D-glucoside?17?,5-methoxy-5-hydroxymethyl-2-furanone?18?,astragaloside?19?respectively.Compounds 7,16,17,18 were frist isolated from Jianpi Shengxue Granules,compounds 18 was reported for the frist time as the natural product.MTT method was adopted to detect the cytotoxicity of the extract from Jianpi Shengxue Granules and the 19 compounds with human normal liver cells HL-7702,to provide certain reference data support for the safety evaluation of Jianpi Shengxue Granules.
