Study On The Pharmacodynamics And Mechanism Of Jianpi Shengxue Tablet

Jianpi Shengxue Tablet have been used for clinic for many years. It plays the role to rise the blood stably. But the research of traditional Chinese medicinel is not exact. This project explore the mechanism of the active ingredient further use pharmacodynamics and mechanism research and lay the foundation to modern research and the secondary development of the Jianpi Shengxue Tablet.Copy iron deficiency anemia in a rat model. After building the model, Jianpi Shengxue Tablet, traditional materia medica and ferrous sulfate were given separately by intragastric, the control group was given the same mount of deionized water. Then measure the index of peripheral hemogram and serum iron 20 days later. The index of peripheral hemogram of the Jianpi Shengxue group was obviously higher than that of others. It shows that the Jianpi Shengxue Tablet, traditional materia medica and ferrous sulfate can treat iron deficiency anemia rats well, but the effect of Jianpi Shengxue Tablet is better than that of the rest of the groups.Copy renal anemia in a rat model. Animals and groups are same as the iron deficiency anemia rats. Then measure the RBC, HGB, HCT, erythropoietin(EPO), blood urea nitrogen(BUN), creatinine(Cr) and osmotic fragility of RBC. The Jianpi Shengxue group, traditional materia medica group and ferrous sulfate group can all improve the content of BUN and Cr without obvious difference. But the Jianpi Shengxue Tablet can obviously increase the EPO, RBC, HGB and HCT. It also dropped osmotic fragility of RBC to the normal range. Jianpi Shengxue Tablet improved renal anemia rats index of peripheral hemogramand reduced the content of BUN, Cr. It shows that Jianpi Shengxue Tablet can treat the renal anemia rats obviously.Copy bone marrow inhibitory anemia in a rat model. Animals and groups are same as the iron deficiency anemia rats. Then detection the content of white blood cells(WBC), RBC and interleukin 2(IL-2). After the treatment, the RCB HGB, HCT of experimental groups are higher than the content of control group. The IL-2 of control group was also significantly higher than the rest of the groups. The Jianpi Shengxue Tablet, traditional materia medicine and ferrous sulfate can treat bone marrow inhibitory anemia model mice well, but the effect of Jianpi Shengxue Tablet is superior to that of the rest of the group.Copy qi deficiency and spleen deficient anemia in a rat model. Animals and groups are same as the iron deficiency anemia rats. The serum hemolysin of mice, expurgation index and phagocytosis activity were significantly higher than that of the rest of the groups. Traditional materia medica and ferrous sulfate have an impact on the above indexes, but not as significantly as the Jianpi Shengxue group. Jianpi Shengxue Tablet can improve the immunologic function with significant efficacy to qi deficiency and spleen deficient anemia model mice.Inspect the effect of Jianpi Shengxue Tablet to the divalent metal transporter proteins(DMT-1)of rats and the expression of Hepcidin gene. The amount of DMT-1 protein expression in intestinal mucosa increase as the concentration rise. The expression of DMT-1 of ferrous sulfate group was obviously decreased compared with model control group. The Hepcidin mRNA expression in the liver increase as the drug dosage rise. The composition of Chinese medicine play an important role to raise DMT-1, due to that, Jianpi Shengxue Tablet have a better effect than ferrous sulfate.Above studies have shown that Jianpi Shengxue Tablet can treat the four kinds of anemia animal models obviously. Jianpi Shengxue Tablet promote the absorption of ferrous iron by the expression of DMT-1. The Hepcidin In the livers effect the absorption of iron by negative feedback mechanism. The composition of Chinese medicine play an efficient role by increasing Hepcidin mRNA expression.
