Bolbostemma paniculatum(Maxim.)Franquet,one of the Chinese folk medicines named as"Tu-bei-mu",is often used for the treatment of tumors and warts as well as for detoxication.Previous research indicated that the main constituents in this plant are triterpenoid saponins,which have anticancer and antivirus bioactivity.On the Based of the review on the chemical constituents and pharmacological research of Bolbostemma paniculatum,the chemical constituents were further studied and their antitumor activities in vitro were tested.By all kinds of chromatographic methods such as silica gel,sephadex LH-20,ODS column and HPLC,37 compounds were isolated from the 75%ethanol extract of the bulbs of Bolbostemma paniculatum.On the basis of spectroscopic analysis(UV,IR,NMR,MS)and physicochemical characters,their chemical structures were identified as tubeimoside D(1),tubeimoside ?(2),tubeimoside ?(3),tubeimoside ?(4),dexylosyltubeimoside ?(S),lobatoside C(6),tubeimoside A(7),tubeimoside B(8),lobatoside A(9),tubeimoside C(10),tubeimoside ?(11),7?,18,20,26-tetrahydroxy-(20S)-dammar-24E-en-3-O-?-Z-(4-acetyl)arabi-nopyranosy-(1?2)-?-D-glucopyranoside(12),isocucurbitacin B(13),23,24-dihydroisocu-curbitacin B(14),cucurbitacin E(15),23,24-dihydrocucurbitacin E(16),(22E,24S)-5?,8?-epidi-oxy-24-methyl-cholesta-6,9(11),22-trien-3?-ol(17),(22E,24S)-5?,8?-epidioxy-24-methyl-chole-sta-6,22-dien-3?-ol(18),?-spinasterol(19),?-sitosterol(20),daucosterol(21),hesperidin(22),kaempferol(23),quercetin(24),emodin(25),physcion(26),vanillic acid(27),p-hydroxybenzoic acid(28),?-methyl pyrrolidone(29),maltol(30),allantoin(31),adenosine(32),acetol ?-D-glucoside(33),gingerglycolipid A(34),5-hydroxymethylfural(35),alanine(36),maltose(37).Among them,1,7-8 and 10 were new compounds;6,9,14,16-19,22-24,27,33-34 were isolated from this genus for the first time.In the course of our search for triterpene saponins with antitumor activity,by MTT methods,the in vitro cytotoxicity activities against cancer cell lines of some compounds isolated were assayed.The effects of the compounds on human cancer cells growth,expressed as the IC50 value.For BGC-823 cells line,compounds 5 was the most potents with the IC50 value of 5.44?M.For A549 cell line,compounds 1,3 and 5 showed strong activities with IC50 value of 8.67,8.82,10.03?M.For HT-29 cell line,compounds 1-7,13 and 16 showed strong activities with IC50 value of 7.48,8.11,4.59,2.10,1.70,6.87,4.18,9.43,2.63?M.For MCF-7 cell line,compounds 1,3-6 and 16 showed strong activities with ICso value of 1.40,3.65,8.54,6.40,8.39,1.20?M.For HeLa cell line,compounds 1-3,5-6,13,15 and 16 showed strong activities with ICso value of 6.86,3.37,8.22,7.68,5.47,6.76,4.47,1.88?M.
