Purpose: To isolate and identificate the saponins from the steams and leaves of panax quinquefolium.Method: On the review of chemical constituents of panax quinquefolium L. and its pharmacological activities, we investigated triterpenoid saponins of panax quinquefolium steams and leaves. Six compounds were isolated from it by using open-column(silica gel,macroreticular resin, Sephadex LH-20, ODS chromatographies), and preparative HPLC methods, etc. On the basis of spectroscopic analysis, including 1H NMR, 13C NMR, ESI-MS and chemical methods, the structures of six compounds were identified.Result: Six compounds were identified as 6-O-α-L-rhamnopyranosyl-(1â†'2)-β-D-glu- copyranosyl-dammar-20(24)-epoxy-3β,6α,12β,25-tetraol (P-F11,â… ); 3-O-β-D-glucopy- ranosyl-(1â†'2)-β-D-glucopyranosyl-20-O-β-D-glucopyranosyl-dammar-24-ene-3β,12β,20S- triol (Rd,â…¡); 3-O-β-D-glucopyranosyl-(1â†'2)-β-D-glucopyranosyl-20-O-α-L-arabino- funosyl-(1â†'6)-β-D-glucopyranosyl-dammar-24-ene-3β,12β,20S-triol (Rc,â…¢); 3-O-β-D- glucopyranosyl-20-O-α-L-arabinofunosyl-(1â†'6)-β-D-glucopyranosyl-dammar-24-ene-3β,12β, 20S-triol (N-Fe,â…£); 3-O-β-D-glucopyranosyl-(1â†'2)-β-D-glucopyranosyl-20-O-β-D- xylopyranosyl-(1â†'6)-β-D-glucopyranosyl-dammar-24-ene-3β,12β,20S-triol (Rb3,â…¤); 3-O-β-D-glucopyranosyl-dammar-24-ene-3β,12β,20R-triol (20(R)-Rh2,â…¥).Conclusion: The purities of compoundsâ… -â…¥were determinated to 100%, 98.1%, 94.6%, 98.4%, 100% and 97.5% respectively by analytical HPLC. Compoundsâ… andâ…¡with high contents and purities were chose as the reference substances. |
