Study On TCM Constitution Types Of Knee Osteoarthritis Patients And LRP5 Gene Polymorphisms

Objective: To explore the relationship among knee osteoarthritis,TCM constintutiontypes and LRP5 gene polymorphism by investigating KOA patients of TCMconstintution types,pathogenesis risk factors and LRP5 gene polymorphism,Providing new ideas and theoretical basis for improving the prevention andtreatment to KOA. Methods: From February 2014 to June 2015,245 patients from Gansu Provincial Hospital of TCM with KOA were investigated by questionnaires,and 128 normal adult volunteers without kinship were randomly assigned as the control samples.ALL of them were provided the informed consents. The main outcome measures including age, gender, ethnicity,body mass index, age, constitution and related risk factors etc.The database was established by Excel,statistical analysis was done by SPSS19.0.The gene polymorphism of LRP5 was detected in some high-frequent types. Results:(1) 245 patients with KOA(32 males and 213 females) and 128 patients with volunteers(71 males and 57 females) were surveyed.(2) There was statistical significance in the initial pain age between city and rural patients from case group, There was no statistical significance in the initial diagnosis age between them(P<0.05).(3) There was statistical significance in body mass index between casegroup and control group(P<0.05).(4) There is statistical significance in the age of menarche, menopausebetween case group and control group of women(P<0.05).(5) There was statistical significance in Occupation Classification, working environment and culture degree between case group and control group(P<0.05).(6) In case group, the incidence of normal type in males was higherthan females,and the difference was statistically significant(P<0.05).(7) The top three biased constitution of KOA are Yang deficiency constitution,Yin deficiency constitution,Qi deficiency constitution;simplybiased constitution of the top three are Yang deficiency constitution,Yin deficiency constitution,Qi deficiency constitution;Participate inpart-folder constitution of the number of the top three are Yangdeficiency constitution,Qi turbid constitution and Qi deficiency constitution.(8) LRP5 gene rs3736228 SNP genotype, allele showed no statistical difference in the results. Conlusion:(1) The risk factors of the incidence of KOA are female gender, age,body mass index, menarche age, menopausal age and TCM constitution of Yang deficiency, yin deficiency and Qi deficiency.(2) The incidence of KOA was associated with Occupation Classification, working environment, cultural level.(3) LRP5 gene rs rs3736228 SNP genotype, allele occurs in KOA hasno correlation.
