Objective To establish the rat model of kidney qi deficiency asthma.To test the level of IFN-??IL-17?IL-6 in broncho-alveolar lavage fluid(BALF).To Observe the difference between disease and syndrome in kidney qi deficiency in asthma.To Analysis the level of IFN-??IL-17?IL-6 in kidney deficiency asthma rats after treatment of Yishen Chuanning Decoction combined with Pulmicort.To study the mechanism of treatment in kidney qi deficiency in asthma by Yishen Chuanning Decoction preliminarily in order to provide experimental basis for clinical.Methods 50 male rats of clean grade were selected and randomly assigned into 5 groups which were the control group(including normal control group,asthma group,kidney qi deficiency asthma group)and the drug group(including Western medicine group,and combination of Chinese and western medicine group).The model was established by asthma combined with kidney qi deficiency syndrome,and the general performance of the rats was observed.The enzyme linked immunosorbent assay(ELISA)was used to test the serum testosterone(T),cortisol(COR),thyroxine(T4)levels.The lung tissues of the rats were stained by hematoxylin-eosin(HE),pathological conditions were observed,and the broncho-alveolar lavage fluid of the rats were obtained to test the cytokines,IFN-??IL-17?IL-6 levels.Results ?There was no inflammation in the rats of normal control group,the shape of trachea and alveolus pulmonis were normal.There were inflammations in the rats of the asthma group and the kidney qi deficiency asthma group compared with the control group,and the shape of the trachea was abnormal,tracheal wall and alveolar wall were thickened.?After established the models,compared with normal control group,the levels of the rat serum COR decrease(P<0.05)in the asthma group,there are no significant differences in T4 and T value in the asthma group between normal control group(P>0.05);the levels of the rat serum T4,COR,and T decrease(P<0.05)in the kidney qi deficiency asthma group comparing to the normal control group;,the levels of the rat T4,COR,and T increased(P<0.05)after receiving treatment in the Western medicine group and combination of Chinese and western medicine group.?After established the models,the value of IFN-? levels decrease(P<0.05)and the value of IL-17,IL-6 levels increased(P<0.05)in the asthma group and the kidney qi deficiency asthma group;the value of IL-17 in the kidney qi deficiency asthma group increase significantly comparing to the asthma group but the value of IL-6 between the asthma group and the kidney qi deficiency asthma group have no significant differences.Comparing to the kidney qi deficiency asthma group the value of IFN-? levels increased(P<0.05)and the value of IL-17,IL-6 levels decreased(P<0.05)after receiving treatment in Western medicine group,and combination of Chinese and western medicine group.Conclusion ?The model of kidney qi deficiency asthma could be established successfully by combined panic stimulation method with ovalbumin sensitized excitation method.?The value of IL-17 in the kidney qi deficiency asthma increase significantly compared to the asthma group.The value of IFN-? and IL-6 have no differences in asthma group and the kidney qi deficiency asthma group,which means that the kidney qi deficiency asthma may be more severe than athma.?Yishen Chuanning Decoction can regulate IFN-??IL-17?IL-6 effectively,but the specific mechanisms remain to be further studied.?It is proved that compared with Pulmicort treatment Yishen Chuanning decoction combined with Pulmicort in treatment of kidney deficiency asthma effect is good,and provided the experimental basis for clinical treatment. |
