Objective Establish the rat model of kidney yang deficiency asthma.By detecting TARC and induction of related cytokines IL-4,IL-5,IL-13 levels,to confirmed TARC is a key link for the pathogenesis of asthma.Through using the methods of molecular biology to test the contents of TARC downstream related cytokines,to verify the efficacy of modified jinkuishenqiwan of treating bronchial asthma,to improve the mechanism of bronchial asthma treatment by modified jinkuishenqiwan,and to explore the details in TARC.Methods1.Based on TCM etiology and Combined with egg albumen sensitization method to establish the rat model with kidney Yang deficiency asthma.2.40 male rats of clean grade were selected and randomly assigned into 4 groups which were control group,kidney yang deficiency asthma group,Chinese medicine group,and western medicine group.The general situation of rats were observaed.The enzyme linked immunosorbent assay(ELIS A)was used to test the serum testosterone(T),cortisol(COR),thyroxine(T4)thymus and activation regulated chemokine(TARC),Immunoglobulin E(IgE)levels.BALF were obtained to test the cytokines,IL-4,IL-5,IL-13,levels and counted White cells.HE-stained was used to observe the changes of pulmonary histopathology.TARC protein in the lung tissue was assessed with immunohistochemistry.3.According to modified jinkuishenqiwan flavored TARC and the influence of related cytokines,explore modified jinkuishenqiwan in the treatment of kidney Yang deficiency asthma.Results1.The model rats with kidney Yang deficiency asthma were were set up successfully.2.Kidney yang deficiency asthma group compared with control group:? There was no inflammation in the rats of the control group,the shape of trachea and alveolus pulmonis were normal.There were inflammations in the rats of the kidney yang deficiency asthma group compared with the control group,and the shape of the trachea was abnormal,tracheal wall and alveolar wall were thickened.?After successfully established the models,the levels of the rat serum T4,COR,and T decreased(P<0.05)in the kidney yang deficiency asthma group;the levels of the rat T4,COR,and T increased(P<0.05)after receiving treatment in the Chinese medicine group,and western medicine group.? After successfully established the models,the value of IL-4,IL-5,IL-131evels increased(P<0.05)in the kidney yang deficiency asthma group;the value of IL-4,IL-5,IL-131evels decreased(P<0.05)after receiving treatment in the kidney yang deficiency asthma group.?After successfully established the models,the value of TARC,IgE levels increased(P<0.05)in the kidney yang deficiency asthma group;the value of TARC,IgE levels decreased(P<0.05)after receiving treatment in the kidney yang deficiency asthma group.? After successfully established the models,the value of TARC gene expression increased in the kidney yang deficiency asthma group,the TARC gene expression level were both significant decreased.Conclusion1.Multi-factors combined egg albumen sensitization in the research to establish kidney Yang deficiency asthma rat model were proved to be stable and reliable.The downtrend of the rat serum T4,COR,T levels of the kidney yang deficiency asthma group suggested that the imbalance of the hypothalamic-pituitary adrenal axis function might be existed in this group.The content of TARC in serum of asthma model of kidney yang deficiency were increased,the positive expression of TARC in lung tissue were increased,the the IL-4?IL-5?IL-13 value levels in the BALF were increased,suggested that the kidney yang deficiency rats with asthma induced by TARC may exist Th2 secretion of IL-4,IL-5,IL-13 increase.2.Modified jinkuishenqiwan had anti-inflammatory action,decreased white blood cells and EOS,repaired of tracheal mucosa and airway mucus,improved the environment of lung inflammation in asthma rats,and this could improve the body hormone level in asthma rats,increase of serum T,COR and T4 levels in hypothalamus.Modified jinkuishenqiwan by regulating the expression of TARC gene,reduced the secretion of IL-4,IL-5,Th2 IL-13,to provide new ideas for clinical treatment of asthma. |
