The Research On Effects Of Compound XingXia Decoction On The Glucose Metabolism Of Phlegmy Wet Type Polycystic Ovary Syndrome Mice

Objective:With the high fat and high glucose food,DHEA and STZ to establish the model of phlegmy wet type PCOS mice,observed the efficacy of compound xingxia decoction on the body weight,ovary morphology,vaginal exfoliated cells,sex hormone,glucose metabolism,leptin and adiponectin of Model Mice,explore the mechanism to the phlegm wet type PCOS,to provide experimental basis of warming the kidney,dissolve sputum and dry wet treatment of traditional Chinese medicine for clinical treatment in phlegm wet type PCOS.Methods:72 female ICR mice of 23 day old were randomly divided into blank group,model group,western medicine and chinese medicine group afte two days adaptive feed,and each group was 18 mice.Blank group:fed normally,25 day old began to subcutaneously inject 0.1 soybean oil ml on the neck and back daily lasted for 34 days,at the same time,lavage with 0.3ml/10g saline water,45 day old injected intraperitoneally with O.lml/lOg sodium citrate buffer once.Model group:fed with the high fat and high glucose food,25 day old began to subcutaneously inject 0.9mg/10g DHEA with the same volume on the neck and back daily lasted for 34 days,at the same time,lavage with 0.3ml/10g saline water,45 day old injected intraperitoneally with 120mg/kg STZ once,injection dose O.lml/lOg.Western medicine group:like model group,use 0.3mg/kg Diane-35 instead of saline water(equal to the adult),and lavage dose was 0.3ml/10g.Chinese medicine group:like model group,use compound xingxia decoction instead of saline water,diluting decoction of 2g/ml into 0.8g/ml(equal to the adult),and lavage dose was 0.3ml/10g.All the mice were executed 2 weeks later after intrapentoneal injection,and collecting specimens.From 28 day old,each group of mice was taken a vaginal smear daily,and observe the change of vaginal exfoliated cells after staining to understand whether oestrous cycle disordered,and record the weight gain and ovarian wet weight,and detect the serum E2,LH,FSH,T,FINS,leptin and adiponectin with ELISA method,and calculate the ovarian index,LH/FSH ratio,HOMA-IR,and observe the change of ovary morphology by HE staining.Results:The vaginal fluid smear of model group was disorder,and ovarian pathological section showsd that ovary had an polycystic change;The estrous cycle of chinese medicine group and western medicine group was basically recovered,and ovarian polycystic change of this two groups was improved,and the improvement of chinese medicine group was more obvious than that of western medicine group.The weight increment,ovarian wet weight,ovarian index,LH,T,LH/FSH ratio,FPG FINS,HOMA-IR,leptin and adiponectin of model group was a significant increase compared with normal mice,with significant difference(P<0.01 or P<0.05);The weight increment,ovarian wet weight,ovarian index,LH,LH/FSH ratio,FPG,FINS,HOMA-IR,leptin and adiponectin of chinese medicine group decreased obviously compared with model group,with significant difference(P<0.01 or P<0.05);The weight increment,ovarian wet weight,LH,LH/FSH ratio,FPG,FINS,HOMA-IR,leptin and adiponectin of western medicine group decreased obviously compared with model group,with significant difference(P<0.01 or P<0.05);The ovarian wet weight,ovarian index,T,FPQ FINS,HOMA-IR and leptin of chinese medicine group decreased compared with western medicine group.Conclusion:Compound xingxia decoction could improve the whole situation of phlegmy wet type PCOS mice by improving insulin resistance,increasing insulin sensitivity,adjusting leptin and adiponectin and adjusting the hypothalamus pituitary gonadal axis.
