Objective: To investigate the relationship between the A/G polymorphism at 77 bp upstream of the transcriptional start of the matrix metalloproteinase-13(MMP-13)gene promoter and the genetic susceptibility of osteoarthritis in Han and Uygur women in Xinjiang Uygur Autonomous Region.METHODS: A total of 802 people with normal knee skeletal arthritis and normal physical examination among Han and Uyghur females in Shihezi,Urumqi,and Nongyi 51 group of residents in Xinjiang Uygur Autonomous Region were surveyed using the current situation investigation method.Blood samples of 5 ml were collected and placed in anticoagulant tubes.There were 334 cases of Uyghur people,93 cases of osteoarthritis group,241 cases of non-osteoarthritis group,468 cases of Han nationality,323 cases of osteoarthritis group,and 145 cases of non-osteoarthritis group.Each group of peripheral blood leukocyte DNA was extracted and polymerase chain reaction(PCR)restriction fragment length polymorphism was used to detect MMP-13A-77 G gene polymorphisms.The osteoarthritis and non-osteogenic features of Han and Uygur patients were compared.Arthritis gene polymorphism distribution differences.Results: The general conditions were compared: Uighur age(57.54±0.35)years,BMI(24.27±0.11)Kg/m2,Han age(54.74±0.30)years,and BMI(24.12±0.09)Kg/m2.The distribution of MMP-13A-77 GSNP genotype of Han and Uygur in Xinjiang Uygur Autonomous Region accords with Hardy-Weinberg equilibrium(P>0.05).The allele and genotype frequencies of MMP-13A-77 G SNP are distributed in the Han and Uygur populations.Significant difference(=31.185,P < 0.001).In the Han population,the gene G is more abundant,and in the Uygur population there are more A genes.The frequency of three genotypes of MMP-13A-77 G SNP in Uygur population was significantly different between osteoarthritis group and non-osteoarthritis group(P<0.001),and compared with AA genotype AG,GG Genotypes significantly reduced the risk of osteoarthritis(?2=0.442 and 0.428;P values ??were less than 0.001),adjusted by age and body mass index(95% CI=0.066-0.371 and 0.090-0.481;OR=0.156 and The frequency of MMP-13A-77 G AA and GG genotypes in the Han population was significantly different between the osteoarthritis group and the non-osteoarthritis group(P<0.001).AG genotype in osteoarthritis There was no significant difference between the non-osteoarthritic group and the non-osteoarthritis group(P=0.691).Compared with the AA genotype,the GG genotype significantly increased the risk of OA(?2=0.447;P<0.001).The index was adjusted(95%CI=2.640-15.213;OR=6.338).Conclusion:1.MMP-13 gene polymorphism is associated with the risk of osteoarthritis.MMP-13 gene is a susceptibility gene for osteoarthritis.2.The down-regulation of genotype AG or the up-regulation of genotype AA in the Han and Uygur genotypes may be more closely related to the risk of osteoarthritis,but genotype GG is associated with a very low incidence of osteoarthritis.3.Age and body mass index are risk factors for osteoarthritis.
