PurposeThis research applys the Network Pharmacology technical methods to explore the mechanism of using Gou-Pi Plaster to cure tension-type headache(TTH).MethodRetrieval the chemical composition of Gou-Pi Plaster components in Traditional Chinese Medicine @Taiwan Database(a Taiwan Traditional Chinese Medicine database),retrieval the the chemical composition corresponding target protein in the PubChem database,and retrieval genes associated with TTH in the Gene database.Upload data to a biological information analysis software of IP A,get the Gou-Pi Plaster and TTH corresponding molecular networks,and then via the IPA(Ingenuity Pathway Analysis)analysis to get the Gou-Pi Plaster and TTH related signaling pathways,finally analyzes the result to explore the molecular mechanism of Gou-Pi Plaster treatment of TTH.ResultRetrieved 351 the chemical composition of Gou-Pi Plaster and 369 target protein altogether;TTH related genes,a total of seven.According to the interactions between molecules,constructed 25 Gou-Pi Plaster molecular networks(composed of 761 nodes)and 1 TTH molecular network(composed of 35 nodes)by IP A software.Further analysis of the IPA scored above 30 points networks of Gou-Pi Plaster merged with TTH network,find there were 16 key interaction target proteins:GAPDH,HIF1A,PKM,ABCC1,ADRB2,AR,DRD2,HTR1B,HTR1D,HTR2A,HTR2B,HTR2C,VDR,ADRA1A,ADRA1D,OPRM1.TTH participates in 77 diseases and biological function and involves in the 51 classical signaling pathways;Gou-Pi Plaster participates in 78 diseases and biological functions and involves in the 390 classical signaling pathways;Gou-Pi Plaster with the participation of TTH are involves in 45 signaling pathways.A comparative analysis of the common participation signaling pathways,the most relevant correlation top 5 respectively are:Serotonin Receptor Signaling is mainly responsible for neuromodulation.LPS/IL-1 Mediated Inhibition of RXR Function(lipopolysaccharide/interleukin-1 mediated by retinoid X receptor inhibition pathway),is mainly responsible for immune regulation;Atherosclerosis Signaling is mainly responsible for disease related regulation;Superpathway of Melatonin Degradation and Xenobiotic Metabolism Signaling are mainly responsible for metabolism.Conclusion(1)Gou-Pi Plaster may regulate neurotransmitters,reduce nerve cells and cerebral vascular injury,relieve the neurogenic inflammation from the neural regulation on treatment of TTH.(2)Gou-Pi Plaster may through immune regulation,and a variety of signal transduction pathways inhibit the release of inflammatory factors,play anti?inflammatory reaction;,which has therapeutic effects on TTH.(3)Gou-Pi Plaster may regulate the lipid metabolism,xenobiotic compounds metabolism and degradation of melatonin metabolism,regulation of cerebral blood supply,antioxidant activity,scavenging free radicals,inhibiting cell apoptosis to protect the nerve,can play a role in the treatment of TTH.
