Objective:To investigate the effects of Chaihushugansan Decoction on the expression of Endoplasmic reticulum stress-related molecules IRE1 and TRAF2 in the Antrum of Rats with Functional Dyspepsia.Methods: We taked the SPF SD rats 30,they were randomly divided into normal control group,model group,saline group,domperidone group and the group of Chaihushugansan,six rats in each group.Rats in the normal group were administrated with normal breeding,model group only clip tail not feeding,while the others were gavaged with corresponding drugs.The concentration of Chaihushugansan and domperidone is 0.22g/ml and 0.21mg/ml,the dose of all rats is 1.5ml/100 g.Observe the rats' changs of life state in the process of building.After 4 weeks of intervention,the gastric emptying of rats were recorded.The histopathological changes of gastric antrum were observed by HE staining.The mRNA level and protein level of IRE1,TRAF2 in the gastric antrum were detected with RT-qPCR and immunohistochemistry respectively.Results: Clip tail stimulus last 4 weeks later,the rats in cluster pull over and don't like activity,aggressive,easily frightened,hair dry bad performance,such as Chaihushugansan and domperidone groups above relatively was not significant.Four weeks after caesarean section in the stomach,no obvious flush swelling or erosion lesions of normal group‘s gastric mucosa,such as residue in model group and saline group,gastric mucosa is a bit flush,Chaihushugansan and domperidone groups the performance was not significant.The HE staining pathological examination showed no obvious inflammatory cell infiltration structure abnormality,no organic lesion.The gastric emptying of model group,saline group decreased compared with normal control group(P<0.05);the gastric emptying of domperidone group,the group of Chaihushugansan increased compared with model group(P<0.05).The mRNA level of IRE1,TRAF2 and protein levels of IRE1,TRAF2 in model group and saline group significantly increased compared with normal control group;The mRNA level of IRE1,TRAF2 and protein levels of IRE1,TRAF2 in domperidone group,the group of Chaihushugansan decreased compared with model group.Conclusion:Chaihushugansan can promote the functional dyspepsia rats stomach power,which may be related to the inhibition of Endoplasmic Reticulum Stress-related Molecules IRE1 and TRAF2.
