Effects Of Chaihushugansan On The PERK/eIF2? Signaling Pathways Of Er Stress In The Antrum Of Rats With Functional Dyspepsia

Objective:Based on the PERK/eIF2? signaling pathway of endplasmic reticulum stress,to investigate the therapeutic mechanism of Chaihushugansan for treating functional dyspepsia(FD)rats.Methods:(1)Totally 24 healthy adult SD rats were randomly and evenly divided into normal group,model group,Chaihushugansan group and domperidone group.(2)The FD rat model was established by tail clamping in all groups except the normal one.From day one,rats in normal group and model group were administered with Saline by gavage,while rats in Chaihushugansan group and domperidone group were treated respectively with Chaihushugansan decoction and domperidone suspension by gavage.All medication lasted for 4 successive weeks.(3)After 4 weeks,stomach tissue of rats in all groups were separated and visually observed.Meanwhile,the histological changes of stomach tissue were observed by HE staining with optical microscope.Gastric emptying of rats was recorded and pathological sections of antrum were detected to appraise the FD model,and utilize immunohistochemistry(IHC)technique to measure the PERK and p-eIF2? levels of stomach-antrum tissue in rats.The relative expression of PERK protein and the ratio between relative expression of p-eIF2? and eIF2? protein were detected by Western blot.Results:(1)More residual contents in the stomach of rats in model group,and there were relatively few residues in the stomach in normal,the Chaihushugansan group,and the domperidone group.(2)The gross and light microscope observation of gastric tissues in all group showed no signs of flushing,swelling,ulcers,metaplasia,and other organic lesions.(3)The gastric emptying rate of rats in each group: Compared with normal group,the gastric emptying rate of model group was significantly decreased(P<0.01).Compared with model group,the gastric emptying rates of rats in ChaihuShugan San group and domperidone group were both increased(all P<0.05).Compared with domperidone group,the rate of gastric emptying in ChaihuShugansan group increased(P<0.05).(4)The expression of PERK and p-eIF2? in gastric antrum of rats in each group was detected by immunohistochemistry: Compared with normal group,the expression of protein PERK and p-eIF2? were increased(P<0.01)in model group.Compared with model group,the expression of protein PERK and p-eIF2? of Chaihushugansan group and domperidone group were increased(P<0.01).Compared with domperidone group,the expression of protein PERK and p-eIF2? were decreased(P<0.05)in Chaihushugansan group.(5)The relative expression of protein PERK and the ratio of relative expression between p-eIF2? and eIF2? protein in the gastric antrum of rats in each groups were detected by Western Blot: Compared with normal group,the relative expression of protein PERK and the ratio of relative expression between p-eIF2? and eIF2? protein was increased(P<0.05)in model group.Compared with the model group,the relative expression of protein PERK and the ratio of relative expression between p-eIF2? and eIF2? protein was decreased(P<0.05)in Chaihushugansan group and domperidone group.Compared with domperidone group,the relative expression of protein PERK and the ratio of relative expression between p-eIF2? and eIF2? protein of Chaihushugansan group was decreased(P<0.05).Conclusion:(1)There might be endoplasmic reticulum stress in gastric tissues of FD rats.(2)Chaishushugansan improved the gastric motility of FD rats may be related to the mechanism of inhibiting the PERK/eIF2? signal pathway of endoplasmic reticulum stress.
