Objective:To observe the effect of deer treasure on gastrointestinal motility and serum NO in functional dyspepsia model rats,and to explore the mechanism,clinical application of this medicine,and provide scientific basis for the promotion of the drug.Methods:60 SD rats from the experimental animal center of Nanjing University of Chinese Medicine,about 160g?220g weight,male and female,to 1 weeks of feeding,divided into 5 groups:control group,model group,domperidone group,Zhang Bao high,low dose group,12 rats in each group,half male and half female.The control group was given normal diet,the other group in the model,the tail.clamp lead to fights joint irregular eating feeding for 14 days.At the end of each model,2 rats were selected to observe the anatomy of rat stomach,duodenum is organic change,to verify the success of modeling.After modeling,the rats of normal control group and feeding.The rats in the model group without treatment,daily only given normal saline,the other model groups were given low dose 90mg/Kg Zhang Bao,Zhang Bao high dose 270mg/Kg and domperidone 2.7mg/Kg solution gavage,on time every day to rats by gavage,the rats for 14 days.After the final dose 24h not feeding(daily drinking water),the detection of the first to be all the rat orbital venous blood 3ml,then each rat to nutritional semisolid paste 0.5mL orally.After 30 minutes,then killed by cervical dislocation.The blood samples under low temperature centrifugation,using enzyme-linked immunosorbent assay method(ELISA)changes in the contents of serum NO detection.The stomach was removed from said its weight,calculation of the rate of gastric emptying,intestine removed at the same time,the measurement of carbon ink length calculation of rat small intestinal propulsion rate.Result:1.The initial rats with similar weight,P>0.05,with random weight standards.At the beginning of the experiment.2.At the end of the model,the stomach and duodenum tissues of 2 rats in each group were observed.There was no organic lesion in the naked eye,which proved the model was successful.3.The model group and blank control group weight,P<0.05;relative were given gavage treatment,domperidone group and Zhang Bao low,average body weight of rats in high dose group were gradually returned to normal,higher than that of the model group(P<0.05).4.Comparison of gastric emptying rate and intestinal drive rate of rats in different groups:?Compared with the blank control group,the gastric emptying and intestinal drive rate of the model group were significantly smaller than those of the blank control group,and the differences were obvious(P<0.05).?Compared with the model group:after the treatment,Zhang Bao group,domperidone group of gastric emptying and small intestinal propulsion rate was significantly higher than that in the model group(P<0.05).5.Detection results of nitric oxide in rats:?Compared with the blank control group,the level of NO in the model group was obviously higher than that in the control group,P<0.01.?Compared with the model group,high,low dose of deer treasure suspension,NO content of domperidone significantly reduced(P<0.05).Conclusion:1.An ideal functional dyspepsia rat model was established by using the complex cause of disease.2.Abnormal gastrointestinal motility and NO secretion may be one of the causes of functional dyspepsia.3.Zhang Bao is one of the effective Chinese medicine in the treatment of functional dyspepsia.4.The possible mechanism of dyspepsia rats roe treasure function is:May and treatment reduce the content of NO,enhance the power of the gastrointestinal tract,so as to solve the FD related symptoms.
