Objective: Research the effect of Chaihushugansan on autophagy within myenteric interstitial cells of cajal(ICC)in functional dyspepsia rats and clarify the mechanism of Chaihushugansan promoting functional dyspepsia motility.Methods: 60 SD rats were randomly divided into normal control group,model group,Chaihushugansan low dose(0.16 g/ml)group,Chaihushugansan medium dose(0.32g/ml)groupand Chaihushugansan high dose(0.64g/ml)group(each n=12).Except the normal control group,the FD model in rats was established by constantly squeezing tails,and then fed the rats with corresponding drugs last for 4 weeks.Measured gastric emptying rate,observed stomach mucosa to the naked eye and stained stomach tissue by HE,surveyed the amount of autophagosome within ICC by transmission electron microscopy,located ICC by immunofluorescence,detected the protein and mRNA expression of Beclin1 and microtubule-associated protein 1 light chain 3B(LC3B)within ICC by immunofluorescence and in situ hybridization.Results: 1.Compared with normal control group,the gastric emptying rateof the rats in the model group was decreased(P < 0.01);compared with the model group,the gastric emptying rate of the rats in the chaihushugansan high dose group and chaihushugansan medium dose group were increased(P < 0.01);compared with the chaihushugansan medium dose group,the gastric emptying rate of the rats in the chaihushugansan high dose group was increased(P < 0.05),the gastric emptying rate of the rats in the chaihushugansan low dose group was decreased(P < 0.05).2.To the naked eye there were no obvious erosion and ulcer of gastric antrum tissue of each group,to the HE staining the gastric mucosa were not being obviously infiltrated by inflammatory cell,etc.3.ICC within normal control group could observe the normal structure of mitochondria,abundant rough endoplasmic reticulum,and occasionally autophagy.ICC within model group and chaihushugansan low dose group could observe abnormal cell structure such as mitochondrial swelling,rough endoplasmic reticulum shed particles and also showing a large number of autophagy;ICC within chaihushugansan high dose group and chaihushugansan medium dose group in the wood cell structure had no obvious abnormalities,mitochondria no obvious swelling,a small amount of rough endoplasmic reticulum degranulation and a small amount of autophagic vacuoles.4.Compared with normal control group,the protein and mRNA expression within ICC of model group were increased(P< 0.01);compared with the model group,the protein and mRNA expression within ICC of chaigaozu and chaizhongzu were decreased(P < 0.01);compared of the chaizhongzu,the protein and mRNA expression within ICC of chaigaozu were decreased(P < 0.05),the protein and mRNA expression within ICC of chaidizu were increased(P < 0.05).Conclusion: 1.The mechanism of gastric motility disorder in FD rats may be related to the over autophagy in the gastric ICC.2.Chaihushugansan maypromote gastric motility in FD rats by inhibiting excessive autophagy of gastric ICC,and its mechanism may be related to down the protein and mRNA expression of Beclin1 then reduce the activity of autophagy and reduced the the protein and mRNA expression of LC3 B and autophagic vacuoles.
