The Study Of Serum Metabolomics In Acute Gouty Joint Model Rats Via Pricking Blood Combined With Medicated Thread Moxibustionon

Objective: Establishing rats model of acute gouty arthritis,and we analyze two groups of serum of rats after treatment of pricking blood combined with medicated thread moxibustion as well as western medicine colchicine.So we can find out the potential biomarkers and deduce their metabolic pathways.Then we can explore the mechanism of function of pricking blood combined with medicated thread moxibustion on serum metabolites in acute gouty arthritis model rats.Method: In this study,39 SD rats were randomly divided into model group,western medicine group and treatment group,13 rats in each group.After modeling,3 rats in each group were randomly selected for HE staining to test whether the model was successful.Rats in Western medicine group were gavaged with colchicine suspension,per 100 g rats body weight were gavaged 1ml as the standard,twice a day,3d continuous.The treatment group took the treatment of pricking blood and medicated thread moxibustion.The pricking blood volume was0.3ml±0.04 ml and thread moxibustion 3 zhuang once a day and continue for 3d.During the treatment,we recorded and analyzed the gait and the joint swelling changes of each group.After the treatment,three groups of rats serum samples were collected,then test the serum metabolite of rats through the means of gas chromatography time-of-flight massspectrometry.At last,Principal Component Analysis(PCA)and Orthogonal Partial Least Squares-Discriminant Analysis(OPLS-DA)were performed.Result:(1)The gait of gout arthritis model rats can be improved significantly with the therapy of pricking blood combined with medicated thread moxibustion.(2)The joint swelling can be alleviated by the therapy of pricking blood combined with medicated thread moxibustion therapy.(3)And there were marked differences metabolites between the serum of the treatment group and the model group rats,such as tyrosine,methionine,alpha-ketoglutarate,xylitoland and so on.These metabolites are involved in the metabolism of citric acid cycle,methionine metabolism,pentose and glucuronic acid conversion,tyrosine and phenylalanine metabolism,etc.And there were marked differences metabolites between the serum of the treatment group and the western medicine group,such as creatine,citric acid,l-malic acid,?-aminopropionic acid,?-ketoglutaric acid and so on.These metabolites are involved in the metabolism of citric acid cycle and pyruvate metabolism.Conclusion:(1)The treatment of acute gouty arthritis by pricking blood combined with medicated thread moxibustion therapy has a obvious effect.Treatment group and western medicine both have equivalent efficacy in improving acute gouty arthritis rats model gait and reducing the swelling degree,but the treatment group is better than the western medicine group in reducing swelling degree.(2)The serum metabolites of rats in the treatment group and the model group were analyzed.And four kinds of signature difference metabolites were found,such as tyrosine,methionine,alpha-ketoglutarate,xylitol and soon.These metabolites can regulate the balance of the metabolism of citric acid cycle,methionine metabolism,pentose and glucuronic acid conversion,tyrosine and phenylalanine metabolism to improve the body's immune function.And there were marked differences metabolites between the serum of the treatment group and the western medicine group,such as creatine,citric acid,l-malic acid,?-aminopropionic acid,?-ketoglutaric acid and so on.These metabolites are involved in the metabolism of citric acid cycle and pyruvate metabolism which can relieve the inflammation and pain of acute gouty arthritis.
