[Objective]Explore the Anti-inflammatory molecular mechanisms of bloodletting therapy's treatment.[Methods]Seventy-eight SD rats were equally divided into Normal group,Bloodletting normal group,Sham operation group,Urate arthritis group,Bloodletting group and Ibuprofen group randomly.By injecting uric acid sodium salt into the right ankle joint cavity so as to set up the acute gouty arthrits model.The Ibuprofen group are treaded by gastric perfusion of ibuprofen,the Bloodletting normal group and Bloodletting group pricked the right Kunlun points.We Measured and recorded the cross section diameter of the injected right ankle joint of the rats before Modeling?before and after the treatment.Observing the morphological changes under light microscope.Assaying mRNA and protein expression levels of IL-1? and TNF-? in rats local ankle by Real-time PCR and Western blot.[Result]According to the result of the cross section diameter,Compared with the Normal group,the cross section diameter in the Urate arthritis group was increased(P<0.05);Compared with the Urate arthritis group and the Ibuprofen group,The cross section diameter in the Bloodletting group was decreased(P<0.05)after the trestment.According to the result of light microscope,a lot of pink crumby substances have been found in the Urate arthritis group with dense inflammation infiltration;small mount of pink crumby substance deposition and inflammation infiltration were in bloodletting group.According to the result of Real-time PCR,Compared with the Normal group,IL-1??TNF-? mRNA expression level in the Urate arthritis group was increased significantly(P<0.01);Compared with the Urate arthritis group,IL-1??TNF-?mRNA expression level in the bloodletting group was decreased(P<0.05).According to the result of Western blot,Compared with the Normal group,IL-1??TNF-? protein expression level in the Urate arthritis group was increased(P<0.05);Compared with the Urate arthritis group,IL-1??TNF-? protein expression level in the bloodletting group was decreased(P<0.05).[Conclusion]The bloodletting therapy for acute gouty arthritis have good therapeutic effect.It can lessen exudate,and improve the Local symptoms in acute gouty arthritis.The expression of IL-1??TNF-? can play a role in anti-inflammatory effects on the bloodletting therapy for acute gouty arthritis.It can be the action mechanism for bloodletting therapy on acute gouty arthrits rats model.
