| As one of the most common diseases in dogs,urinary system diseases cause a high damage rate to the bottom tip of lower urinary tract.In this way,the prescription diet is widely used in the clinical treatment for dogs with urinary diseases,which can be combined with medicine as a supplementary diet and prevent the recurrence of disease.In order to investigate the auxiliary therapeutic effect of traditional Chinese medicine prescription diet on the lower urinary tract injury in dogs,this work established damage models of the urinary crystal and the cystitis on dogs’lower urinary tract,respectively,through the melamine method and the alcohol bladder perfusion method.24 beagles were randomly divided into the control group(6 dogs),the prescription group(6 dogs)and the modeling group(12 dogs).Dogs were fed with regular diet in both the control and the modeling groups,and with prescription diet in the prescription group.Meanwhile,melamine at the amount of 0.6 g/(kg.d)was added to the diet for dogs in the control group and the prescription groups.During this procedure,both blood and urine samples of dogs were regularly collected for laboratory analysis and their clinical symptoms were recorded as well.In the prevention experiment of cystitis,dogs in the original control group(6 dogs)were kept,and the rest(18 dogs)were randomly divided again into the prevention group(6dogs)and the cystitis group(12 dogs).To build the cystitis model,dogs in both the prevention and the modeling groups were conducted with bladder perfusion by using75vol%medical alcohol at 0.6 ml/kg,respectively on the 0thday and the 7thday.In this period,dogs were fed with regular diet in both the control and the modeling groups,and with prescription food in the prevention group.In the treatment experiment of cystitis,dogs in the original control group(6 dogs)were still kept,and dogs in the cystitis group(12 dogs)were further divided into the natural recovery group(6 dogs)and the treatment group(6 dogs).Within two weeks,dogs were fed with regular diet in both the control and the natural recovery groups,and with prescription food in the treatment group.During this procedure,blood and urine samples of dogs were daily collected for laboratory analysis,and their clinical symptoms were recorded again.These results indicated that:1.After fed with melamine,an increase on the WBC and the Gran levels were found for dogs in both the modeling and the prevention groups until being stabilized on the 56thday.Those values reached to their maximum on the 28thday for the modeling group and on the 42ndday for the prevention group.No observable change to the levels of RBC and HGB were found for dogs in both groups.A significant increase and the urea nitrogen levels were found for dogs in both groups on the 28thday,while their values turned to decrease on the 42ndday in the prevention group and on the 56thday in the modeling group.On the 28thday,the occult blood showed a positive(+)result in the modeling group and a minor positive(0+-)result in the prevention group.Centrifuged with the same amount of urine,more crystals were found in the modeling group than the prevention group.On the 42ndday,there showed a significant improvement of the serum phosphorus and magnesium levels in the modeling group along with positive(+)results of urine protein and WBC,but resulted in no significant change in the prevention group.2.In the prevention experiment of cystitis,a continuous increase on both the WBC and the Gran levels was found for dogs in the cystitis group within two weeks,while the increase was evened to their original levels for dogs in the prevention group on the 14thday.The RBC and the HGB levels were found to decline continuously for dogs in the cystitis group,while there showed no significant difference in the prevention group.For dogs in the cystitis group,the serum creatinine and urea nitrogen levels were found to increase significantly,accompanied with a increase on the serum phosphorus level as well.The activities of TNF-αand IL-6 were significantly improved for dogs in the cystitis group on the 7thday,while only the activity of the IL-6 was significantly improved for dogs in the prevention group on the14thday.For dogs in both the cystitis and the prevention groups,there showed a decrease of C3 level and a increase of CRP level,positive(+)results of urinary occult blood,urinary protein and WBC,and a large amount of WBC and bladder epithelial cells were found in urinary sediments.3.In the treatment phase of cystitis,the values of WBC,Gran,RBC and HGB for dogs in the treatment group were found to recover to the same level as those in the control group.Among them,the former two values(WBC and Gran)in the natural recovery group were still significantly higher(P<0.05)than those in the control group,while the latter two values(RBC and HGB)were significantly lower(P<0.05).The CRP level for dogs in both the natural recovery and the prevention groups decreased to the normal level.On the 21stday,the serum magnesium level in both the natural recovery and the prevention groups obviously decreased,while the IL-6activity in the natural recovery group was still obviously higher than that in the control group(P<0.05).In conclusion,the damage of urethritis to the lower urinary tract was greater than that of urinary crystal,and the development of inflammation was faster.The prescription of traditional Chinese medicine could regulate the levels of TNF-α、IL-6and other cytokines. |
