Patellar dislocation is a common disease in dogs,which seriously affects the normal life of dogs.Because of its low cost,simple operation,wide indications and obvious effect,trochlear groove reconstruction has been widely used in the treatment of patellar dislocation.However,the operation of destruction of cartilage layer seriously affects the recovery of dogs after operation,which will cause excessive pain response and postoperative infection.Therefore,the reduction of inflammation and pain response and the promotion of cartilage repair are the aspects that need to be optimized.Platelet-rich plasma technique is a technique to prepare high concentration platelet plasma by centrifugation of autogenous blood.Platelets in plasma can play a role in tissue repair,inflammation and immune response.PRP as a self-produced extract to avoid rejection,has the advantages of rich sources,simple operation and low price,which has a great application prospect in animal therapy.In this study,PRP was injected into the knee after trochlear groove reconstruction.To explore the possibility of PRP preparation in veterinary,and to verify that PRP plays an active role in reducing inflammatory pain,preventing infection and promoting cartilage repair after trochlear groove reconstruction.The content of test:(1)PRP preparation and composition analysis:Platelet rich plasma was prepared by single gel centrifugation.The whole blood and PRP were counted by blood routine to count platelets and leukocytes,and the enrichment coefficient and recovery rate were calculated to verify whether the PRP prepared by this method met the standard and whether it could be effectively used in veterinary.(2)The application of PRP in the repair of injury after trochlear reconstruction in dogs:PRP was injected into the knee joint once a week for 3 weeks.The knee joint was scored 7 days before operation and 5 days after operation.The inflammatory cells in blood were counted.The imaging ultrasound and CT were performed 4 weeks after operation.The gross anatomy and histology were observed and scored 12 and 16 weeks after operation.To verify whether PRP can control inflammation,reduce pain and promote cartilage repair after trochlear reconstruction.The results showed:(1)The experimental dogs took 3 mL of whole blood,the ratio of whole blood to anticoagulant 6:1,and centrifugation 5 min at medium speed of 3600 rpm in the gel anticoagulant tube.The upper serum was extracted,and 5 min was concussion concussion,and 0.2 ml PRP was prepared.The concentration of platelet in PRP was 1532.08±215.29×1O9/L,the enrichment coefficient was 6.08±0.53 and PRP met the requirements of PRP.(2)The application of PRP in the repair of injury after the reconstruction of trochlear groove in dogs:results of knee joint score:compared with the control group,there was significant difference between the experimental group and the control group(p<0.05),and the score of each time point was lower than that of the control group;the leukocyte count and neutrophil count were detected by routine blood test in the laboratory,and the value of tl-t5 in the experimental group was significantly different from that in the control group(p<0.05),and the value of each time point was lower than that in the control group;imaging examination:after 4 weeks of high-frequency ultrasound and CT observation,the continuity of the cartilage surface in the experimental group was better than that in the control group;general observation At the same time,the experimental group and the control group had different degrees of wear and subchondral hemorrhage compared with the normal cartilage tissue,and the experimental group was better than the control group.There was significant difference between the experimental group and the control group in the scores of 12 weeks and 16 weeks(p<0.05).Histological observation score:at the same time point,the experimental group was better than the control group in cartilage thickness,matrix staining and chondrocyte number.There was significant difference between the experimental group and the control group in the score of cartilage tissue at 12 weeks and 16 weeks(p<0.05).The score of experimental group was lower than that of cartilage tissue,and there was significant difference between the experimental group and normal cartilage at 16 weeks(p<0.05)).Comprehensive test results:(1)PRP was extracted from autologous blood by single gel centrifugation.The platelet concentration coefficient was 6.08±0.53,which accorded with the effective concentration.Because its preparation method is simple and the amount of blood drawn can meet the clinical needs,it has a great application prospect in pet clinic.(2)PRP was injected into the knee after trochlear groove reconstruction in dogs.The time interval was one week and lasted for three weeks.It had significant effect on reducing inflammation,controlling pain and promoting cartilage repair.
