| Hyriopsis schlegelii belongs to Lamellibranchia Class,Phylum Mollusca.originated in Japan,with the characteristics of large body,thick shell,well-developed organization and strong ability to secrete pearl.Their flesh quality is often discarded and not utilized when pearls are taken out,.In order to improve the utilization rate of resources of the Hyriopsis schlegelii,the meat quality of mature individuals were selected as the material and its basic nutrients were analyzed.The fresh organs contained water 89.68%,crude protein 5.63%,ash 1.93%,glycogen 1.24%,crude fat 0.96% and crude fat 0.56%.Three proteases(papaya,animal,neutral)were first time used to digest all the organs of the Hyriopsis schlegelii,and the related process was established to obtain three enzymatic products.The basic nutrients and 18 amino acid contents of the enzymatic hydrolysate were detected and analyzed.In vitro antioxidant experiments were designed to investigate the antioxidant capacity of three enzymatic products in vitro by using DPPH,hydroxyl radical and superoxide radical scavenging rates as indicators.The study found that all three enzymatic hydrolysates have the ability to scavenge the above indicators,and the scavenging effect is enhanced with the increase of the concentration of the three enzymatic products.The neutral protease digestion product had the best clearance rate for the three indicators.Taking 0.6 mg/ml as an example,the DPPH,hydroxyl radical and superoxide radical scavenging rates were 23.8%,37.2%,and 19.6%,respectively.Studies have shown that the enzymatic hydrolysate of the Hyriopsis schlegelii has the ability to resist oxidation in vitro.The experimental design of in vitro ADH activation rate,mouse drunkenness test,mouse acute alcoholic liver injury experiment,explored the three enzymatic hydrolysates to protect the biological activity of hangover and alcohol-induced liver injury.The results of in vitro ADH activation rate showed a positive correlation in the concentration range of 3.75-1550mg/mL;the results of drunkenness test showed that the neutral protease digestion product can prolong the tolerance time by 1.5 hours and shorten the intoxication time by 3 hours.There was a significant difference between the two times(p<0.01);the animal protein hydrolysate product had a significant tolerance of 0.5 hours and a shortened intoxication time of 2 hours compared with the model group(p<0.01);papainase There was no significant difference in the tolerance and intoxication time between the product group and the model group(p>0.05).The results showed that animal and neutral protease digestion products have certain effects on hangover.The results of acute liver injury test showed that the enzymatic hydrolysate of neutral protease digestion product significantly increased the content of TG and GSH in liver(p<0.01)and decreased the content of ALT and AST in serum at 150mg/kg BW dose(p<0.01).p<0.05),the low and medium concentration groups were not significantly compared with the model group(p>0.05).The results showed that the enzymatic hydrolysate of Cyprinus sinensis had a certain protective effect on alcohol-induced acute alcoholic liver injury in mice.This experiment explored the anti-tumor and repair of liver damage by extracting the Hyriopsis schlegelii.The SMCC-7721 and SGC-7901 cell lines were used for culture.It was found that HSP-1 promoted the cells at low concentration(100-200 μg/m L),but inhibited at 400 μg/m L,1000 μg/m L.The inhibition rate reached the maximum,which was 12% and 11% for the two cells,respectively,and the inhibition effect was not significant.In the study of alcohol-induced liver injury protection,HSP-1 at 250 mg/kg BW can significantly reduce alcohol-induced acute liver injury and non-acute(4 weeks)liver injury in mice serum alanine aminotransferase(ALT)and aspartate aminotransferase(AST)content(p<0.01),reduced triglyceride(TG)and increased glutathione(GSH)content in homogenate(p<0.05);liver homogenate in non-acute(4 weeks)liver injury mice Although the content of MDA was not significantly decreased(p>0.05),it had a certain trend.The above experimental results indicate that HSP-1 has a protective effect on alcohol-induced liver injury. |
