In the basis of previously studies on plant extract compounds is limited and their mechanism is still uncertain,the present study was to develop new compounds with lower cost and multi-function such as anti-microbial,anti-oxidation and anti-inflammation,and provided scientific basis in animal production.The main results were follows.1.Single kind of plant aqueous extracts in vitro experimentsTwenty-four plants were selected to prepare the aqueous extracts containing crude drug 0.5 mg/?l.The results showed that the inhibitory ratios against Escherichia coli of Punica granatum,Artemisia Apiace,a,Cassia twig,Cinnamon cassia Presl all above 50%.The inhibition rate to 5-lipoxygenase(5-LOX)activity of Punica granatum,Caulis Spatholobi,Magnolia officinalis,Citrus limon,Rosmarinus officinalis all above 90%.And the minimal half concentration of scavenging DPPH radical were Punica granatum(0.08mg/ml),Rosmarinus officinalis(0.31mg/ml),Cortex fraxini(0.71 mg/ml),the other extracts ranged from 1.04 mg/ml to 199.27 mg/ml.2.Study on the combination of plant aqueous extractsAccording to the above results and cost,five plants,like Punica granatum,as the material of compound plant aqueous extracts,through orthogonal design three advantage compounds were received,which were C1(main function was anti-microbial),C2((main function was anti-inflammation),C3((main function was anti-oxidation).The results showed that the minimum inhibitory concentration(MIC)of C1,C2,C3 were 125 mg/ml.The half concentration of inhibiting 5-LOX activity were 0.434 mg/mL,0.138 mg/mL,0.166 mg/ml.And the half concentration of scavenging DPPH radical were 0.31 mg/mL,0.40 mg/mL,0.22 mg/ml.3.Study on safe application dose of plant aqueous extracts compound78 Balb/c female mice were divided into 4 groups containing the control,C1,C2,C3.Except the control every group contained four doses(125,250,500,1000 mg/kg BW),the experimental period was 21 days.The results indicated that all of the mice were maintain a good health,there were no negative effects on growth performance in low dose group of C1 and C2,while growth performance was decreased in high dose group of C3.The indexes of blood routine and viscera showed that in normal range,the viscera indexes of C1 and C2 group was increased,white blood count(WBC)of C1 and C2 in less dose were extremely significant higher than those in control group(P<0.01),the viscera indexes and WBC was no significant differences in C3 groups(P<0.05).4.Study on growth promoting effects of plant aqueous extracts compoundBased on the results of safe application dose test,the following experiment selected Cl and C2 to continue.90 Balb/c female mice(15g)were divided into 3 groups containing the control,C1(added 0.6mg/ml C1 into water),C2(added 0.3mg/ml C2 into water),every group had three repetition,the experimental period was 21 days.The results indicated that compared to control,average daily gain enhanced 13.3%in C1 and C2(P<0.01).5.Study on intestinal function of plant aqueous extracts compound60 Balb/c female mice were randomly divided into 4 groups containing the control,LPS model,LPS+C1(added 0.6mg/ml C1 into water),LPS+C2(added 0.3mg/ml C2 into water),the mice in the control group received once intraperitoneal injections of saline in the 21 day,the other groups were injected with same volume of LPS(10mg/kg),the mice were sacrificed after 4 hours(8 mice)and 24 hours(7 mice).The results of 4 hours after intraperitoneal injection indicated that compared with the control group,the crypt depth(CD)was extremely increased(P<0.01)and the ratio of villus height to the depth of crypt(VH/CD)was extremeely decreased(P<0.01)in jejunum of the LPS model group.In LPS+C1 and LPS+C2 the ratio of villus height to the depth of crypt was extremely higher than those in LPS model group(P<0.01).Compared with the LPS model group,the jejunum interleukin-6(IL-6)expression of LPS + C1 group was extremely decreased(P<0.01),the LPS + C2 group of interleukin-1 ?(IL-1?)and IL-6 expressions were reduced 40.1%and 60.5%(P<0.01).The results of 24 hours indicated that compared with the LPS model group,the villus height(VH)was extremely increased(P<0.01),VH/CD in jejunum was significantly increased of LPS+C1 group(P<0.05),the jejunum VH in LPS+C2 was extremely increased(P<0.01).The expressions of Toll-like receptor 4(TLR4)in LPS+C1 group was extremely decreased(P<0.01),the expressions of IL-10,IL-6,TNF-? and TLR4 in LPS+C2 group were fell by 52.3%?59.5%?46.3%?57.9%(P<0.01).In conclusion,the present study developed two safe and efficiency plant extract compounds which could promoted growth through enhanced immunity,decreased inflammation and protected intestinal function.
