| Hyriopsis schlegelii, which belongs to Hyriopsis Genus, Unionidae Family,Lamellibranchia Class, Phylum Mollusca, is native to Lake Biwa of Japan. FuzhouHongmen Reservoir Exploitation Corporation has introduced initially this musselfrom Japan to China since1997and achieved artificial cultivation propagationsuccessfully in1998. As a different species from Hyriopsis cumingii, Hyriopsisschlegelii has been extensively applied in pearl-producing industry in Japan recentyears for its high disease resistance and excellent quality of produced pearls andshows high economic value. Besides producing pearls, the nacreous mussel shellscan be used for inlaying pearl furniture, ornaments, kitchen utensils and souvenirs..Polysaccharide, which is a kind of macromolecule polymer, is made up ofaldoses and ketoses by glucosidic bond and has complex structure and enormousmolecular weight. It extensively exists in animals, plants and microorganisms and isone of the primary substances keeping vital movement normal. Recent glycobiologystudies show that natural polysaccharides have some special biological activitiessuch as inoxidizability, anti-hypertensive, antibacterial, anti-tumor,anti-inflammatory, hepatoprotective activities and so on.In this study, we extracted polysaccharide from Hyriopsis schlegelii by hotwater and investigated extraction efficiency. The result show that the best extractioncondition is70℃,6hours, H2O: homogenate=25:1v/w. The extraction effiency is27%. The crude polysaccharide solution was then separated through a columnpacked with DEAE-cellulose52. The column was stepwise eluted with pure waterand0.5M sodium chloride solutions. The polysaccharide eluted by water wasconcentrated and purified through a column packed with Sephacry S-300particlesand then determined by the phenol-sulfuric acid method. Elution with higherconcentration was concentrated and the average polysaccharide molecular weightwas2204Da measured by high-performance gel-permeation chromatography.According to GC-MS analysis, the polysaccharide was consists of glucose, arabinose and glucuronic acid. The total contents of glucose, arabinose andglucuronic acid were64.8%,17.4%,17.8%, respectively and glucose is the mainlycomponent. The FTIR spectra of the fraction show the specific absorb peak ofpolysaccharide.In this experiment, we extracted, purified a kind of polysaccharide fromHyriopsis schlegelii and analysis its component which provide a reference forHyriopsis schlegelii polysaccharides research. |
