| In this study, the Cross antigens between Trichinella spiralis and A549Lungcancer cell were determined by ELISA and Western–blotting. Five gene proteineswhich related to A549cells was obtained at immunoscreening from Trichinellaspiralis cDNA expression library, and the antibody anti-tumor effects by animalstudies.Preparation of antigen: First preparation of anti-Trichinella spiralis solubleprotein hyperimmune sera, and anti-A549Lung cancer cell antigen sera, respectively.The cross-reactivity between Trichinella spiralis antigen and A549cell antisera orbetween A549cell antigen and Trichinella spiralis antisera were measured byELISA.The results showed that there are cross-reaction between Trichinella spiralisand A549cells, existed associated antigen. Then the immune response betweenTrichinella spiralis antisera and A549lung cancer cell antigen was detected bywestern-blotting. Three specific reaction bands were obtained, And the reactionantigen presented near9kDa,27kDa,54kDa.Screening A549tumor-related antigen gene from Trichinella spiralis:Associated antigen antisera as screening antibody, immunoscreening Trichinellaspiralis cDNA expression library for positive plaques. Then the phagethe plaquesDNA were extracted, the inserted DNA fragments were been amplified, then the genewere sequencing and bioinformatics analyzing.The gene (Tsp05904, Tsp10855, Tsp06172, Tsp08404, Tsp05091)wascoloned and expressed in E.coli, the the purified protein was use to immunize themice for the antisera.Observation of the anti-tumor effect on the nude mice which was treated by TheCross antigens antisera: The nude mice with lung cancer solid tumor model were eatablished by inoculated with the fresh alive A549cells,20d later, the tumor-bearingnude mice were treated with sera for two weeks, after that, the changes of the tumorsize and weight in mice. Two weeks after treatment, the tumor weight and volumewere calculated. The results show that the antiserum of A549associated antigenproteins has anti-tumor effec(t53.71%,64.77%,52.39%,40.93%,54.63%), Greaterthan the control group (36.44%). These results provided the avidence of researchingTrichinella spiralis anti-tumor effect from different aspects. |
