Griflola frondosa possesses a great development value as a precious food and medicinal fungus. It is reported that polysaccharides of Grifola frondosa have significant anti-cancer function and the preparation has been approved for clinical trials. This research collected 30 Grifola frondosa varieties, screened all the varieties to select high polysaccharides variety, and optimized its growth medium, and studied on the anti-tumor function, the main results are as follows:1. We carried out the research on Grifola frondosa polysaccharides extraction by microwave and hot water method, screened all the varieties to select the highest polysaccharide "GF29", whose polysaccharide content from mycelia was 8.83%,1.50 times of the popularized variety "SM52"(5.86%) and 13.79 times of the lowest polysaccharide variety "GF01"(13.79%).2. In this study, we determined the best economic formula medium for the fermentation of the high polysaccharide variety. Using the orthogonal test and single-factor experiment, the optimal mode is:cornmeal 30 g/L, soymeal 0.3 g/L, KH2PO4 0.2 g/L, MgSO4·7H2O 0.05 g/L, VB1 0.005 g/L. The mycelial yield reached 0.2079 g/100 mL after 10days of incubation at 25℃. With the national average market prices(Corn 1.8 yuan/kg, Potatoes 3 yuan/kg), the high polysaccharide "GF29" produced the same amount of mycelium and the cost formula is only 11.11% of ordinary formulas.3. Through setting up mouse S180 tumor-bearing models, we carried out the research on the anti-tumor functions of the Grifola frondosa polysaccharide. High dosage group's tumor suppression rate was 41.14%(anticancer drug Cyclophosphamide 56.96%), comparing with the varieties "SM52" (23.86%) relative to the 72.42%, and the effect was proportional to the dose. Comparing with the control group, Grifola frondosa polysaccharide (2.43) could obviously enhanced the S180 tumor-bearing mouse's thymus index by 37.28% while the spreading variety "SM52" increased only 16.83%; Spleen index (6.13) increased 36.83% compared to the control group while the variety "SM52" increased only 21.87%.
