Research Of Prescription Of Cultivating Materials Of Tremella And Process Optimization On The Extraction Of Its Polysaccharide

Tremella is one kinds of edible mushrooms,which includeshigh medical value and benefits for human health.With the rapid development of edible mushroom industry,the requirement of tremella had been enhanced to a higher level,especially for the requirement of the active ingredients of the tremella.But there without cultivation standard of planting tremella,most of prescription of cultivating materials that be chosen by experience.According to this situation,the uneven quantity of tremellas cannot meets requirements of customs,especially to the company which produces the health care product.This study pay attention to the prescription of cultivating mater ials of tremella,the standard of quality evaluation of tremella and processing optimization on the extraction of tremella polysaccharide.And it also focuses on finding out the critical influences of the extraction of tremella polysaccharide.Conclusions were drawn as follows:(1)According to adjust the types and ratios of cultivating materials,had found four prescriptions which are meeting different requirement includes the fastest growing speed,the highest fresh weight,the highest dry weight,having highest content of tremella polysaccharide.The detail of the prescription:(1)Meeting requirement of getting the highest growing speed includes 75.00 g of cottonseed hull,55.00 g of jagged wood of reishi,30.00 g hull of sachalnchi,10.00 g of bran,0.50 g of gypsum and 0.10 g of monopotassium phosphate;(2)Meeting requirement of getting the highest fresh weight includes 54.02 g of cottonseed hull,15.43 g of jagged wood of reishi,41.68 g hull of sachalnchi,1.68 g of bran,3.00 g of gypsum and 0.10 g of monopotassium phosphate;(3)Meeting requirement of getting the highest dry weight includes84.95 g of cottonseed hull,74.93 g of jagged wood of reishi,60.00 g hull of sachalnchi,28.14 g of bran,0.15 g of gypsum,0.40 g of monopotassium phosphate;(4)Meeting requirement of getting the highest content of tremella polysaccharide includes 55.00 g of cottonseed hull,15.00 g of jagged wood of reishi,40.00 g hull of sachalnchi,5.00 g of bran,2.50 g of gypsum and 0.20 g of monopotassium phosphate;(2)Prescription of cultivating materials affect the quality of tremella.Firstly,there without the difference of smell,color,and size,but broken rate shown different.Secondly,the content of heavy metal in the tremella are different to each other.Thirdly,themoisture content,protein content,and polysaccharide content are both different,but the ash content is similar.Finally,the rigidity and viscosity of the extracting solution are both different.(3)There are without any correlation between tremella rigidity and its content of polysaccharide,but there is strong correlation between the polysaccharide content and the viscosity of the extracting solution.(4)The optimize condition on the extraction process is,sifter mess is 40,extraction temperature of 90℃,extraction time of 4h and the liquid-to-solid ratio is 60:1(v:w).According to the orthogonal experiment,the effect levels of these factors successively were extraction time,liquid-to-solid ratio,and extraction temperature.
