This paper studied the quality of Tremella’s fruit body cultivated respectively with JUNCAO, cotton seed shell and basswood, contemporarily carried on the comparative ananysis of their quality, for the purpose of scientific assessment of the quality of Tremella’s fruit body cultivated with JUNCAO, and offering scientific basis for the development of JUNCAO Tremella industry. At the same time, research on the polysaccharide and polysaccharide peptide of JUNCAO Tremella was carried out, in order to lay the foundation for the development of the JUNCAO Tremella care products. Results of the specific research were as follows:1. This test checked the content of gossypol, heavy metals and pesticide residues existed in the Tremella cultivated respectively with JUNCAO and cotton seed shell, and comparative studies were also conducted. The results showed that JUNCAO Tremella did not have the gossypol problems (It was not detected). However, a small amount of Gossypol existed in the Tremella cultivated with the cotton seed shell. But the content did not exceed the national standard(as edible oil agricultural industry standard NY/T 751-2007 for the reference standard). The contents of heavy metal and pesticide residues existed in JUNCAO Tremella are all in the scope setted by relevant national standards of Tremella, Green Food standards and the international food standards.2. The test compared and analyzed the main nutrients of Tremella’s fruit body cultivated respectively with JUNCAO, cotton seed shell and basswood, the analysis of Tremella quality showed that:bubble rate, content of ash, moisture, crude fiber and crude protein of Tremella cultivated respectively with JUNCAO, cotton seed shell and basswood were all in the scope setted by the Tremella agriculture industry standard NY/T 834-2004. The ash content of JUNCAO Tremella was 6.44%, crude fiber 1.67%, crude fat 2.14%, crude protein 14.42% crude polysaccharide 31.69%, amino acids 8.65%, higher than Tremella cultivated respectively with the cotton seed shell and basswood. the total sugar content of 69.58%, slightly lower than Tremella cultivated with the cotton seed shell and basswood.3. This test was carried to study on the optimization of JUNCAO Tremella’s polysaccharide extraction conditions by the hot water extraction.Results showed that:the temperature was the most critical factor of those which affected JUNCAO Tremella polysaccharide yield. The optimum extraction conditions were:extraction temperature of 100℃, extraction time of 5 h, the solid-liquid ratio:1:80. JUNCAO Tremella’s polysaccharide yield was the highest under these conditions, extraction rate was 31.29%, 34.78% higher than Tremella cultivated with the cotton seed shell,20.65% higher than Tremella cultivated with basswood.4. This test checked the GA content of polysaccharide of different mass concentrations extracted from the JUNCAO Tremella by Alprenolol Hydrochloride method, to get the optimum scope of mass concentrations of polysaccharide extracted from the JUNCAO Tremella.This scope was 0.3 mg/mL-0.5 mg/mL.Meanwhile, this test was carried on the studies on the GA content of polysaccharide extracted from the Tremella cultivated by different culture medium. The results showed that the GA content of polysaccharide extracted from the Tremella cultivated with fresh JUNCAO was the higest of all the Tremella cultivated with JUNCAO, at the same time, compare respectively with the polysaccharide of Tremella cultivated with cotton seed shell and basswood. There were small difference. It illustrated that the nutritional value of JUNCAO Tremella was not lower than Tremella respectively cultivated with cotton seed shell and basswood.5. Removal of protein existed in polysaccharide of Tremella cultivated respectively with JUNCAO and cotton by using Sevag method. After protein removal of 30 times, there were not protein after checking the proten’s content by the Brandford method. Nextly, by the automatic amino acid analysis, The results showed that polysaccharide of Tremella still contained 17 kinds of amino acids, and Tremella’s polysaccharide combined with the shot peptide and the combination was fairly solid, so the Tremella’s polysaccharide peptide was named.6. Polysaccharide peptide of Tremella cultivated respectively with JUNCAO, cotton seed shell and basswood was preliminarily separated and purified, then furtherly purified and classified by DEAE-Sepharose FF column chromatography. The experimental results showed that:two components of TPP1 and TPP2 were got after DEAE-Sepharose FF column chromatography classification. TPP2 was furtherly purified and identified the purity by Sephadex G-200 molecular sieve gel exclusion chromatography. It was found that TPP2 was the single polysaccharide peptide components. It explained that TPP2 was pure.
