Nowadays,companion animals play an increasingly important role in our life,and pet dog is one of the most common companion animals.A large number of research and clinical practice shows that blood transfusion has a good effect when treating dogs with some fatal infectious diseases(such as Canine Parvovirus、Canine Distemper),acute blood loss,or anemic diseases.In some countries in Europe and America,animal blood bank has already developed a relatively mature system,but there is no standardized animal blood bank in C hina yet.Accoringly,it is important to establish a standardized animal blood bank in the small animal clinical in our country.The aim of this research is to establish the animal blood bank.In this study,the leukocyte is filter out from the whole blood of beagle dogs and is compared with the control group to study the effects of leukocyte remove on blood preservation and transfusion.We first studied the effect of leukocyte remove on blood preservation in beagle blood.Compared with whole blood group,the leukocytes of the experimental group were filtered.The pH,K~+,Na~+,C l~-,FHb and ATP levels of both groups were measured on day 1,day 7,day 14 and day 28,and were to assess the effect of leukocyte remove on blood preservat ion.Then,in order to further study the effect of the leukoreduced blood transfusion,a blood transfusion experiment was carried out in 24 beagles.The body temperature,heart rate and breath were measured 0.5 hours before blood transfusion and 0.5h,1h,2h,4h,8h after transfusion,and 4mL blood samples were collected after each treatment.During the experiment,the dogs were observed to confirm having no abnormal symptoms such as vomiting and shivering.Combined with body temperature,heart rate,respiratory data,whether the transfusion reaction having occurred is determined.The sera were then isolated and the changes of IBIL,ALT and CK is determined to assess the influence of the blood transfusion on organ burden and study the effect of leukocyte re move on blood transfusion.In vitro test results showed that with the prolonging of blood storage time,the concentration of K~+and free hemoglobin in plasma increased,the concentration of C l~-decreased,and the concentration of Na~+fluctuated in a certain range.O n the first day,the content of free hemoglobin in the experimental group was significantly higher than that in the whole blood control group.On the 28th day,the content of free hemoglobin in the whole blood control group was significantly higher than that in the experimental group.There was no significant difference in the risk of transfusion between the experimental group and whole blood group.The concentrations of IBIL,ALT and CK were compared at different time points in each group,and the difference is not significant.Compared with the experimental group and control group,there was no significant difference in the hemolysis of red blood cells and the burden of heart,liver,etc.In summary,compared with whole blood,blood samples with leukocyte remove are more suitable for long-term preservation;and can effectively reduce the incidence of transfusion reaction.Accordingly,it is worth popularizing in small animal clinics.
