Blood transfusion was an important first aid and treatment method in veterinary clinical, and it was widely applied. But the homologous transfusion leads easily to cross-infection of animal diseases. The source of bloodstream was scarcely. The blood is preserved difficultly. It cost many to preserve blood, and the storage period of blood is short. There isn’t special blood bank established for animals in China. It became a research hotspot in the world-wide that researchers look for a safe and effective oxygen carrier. Lecithin was used to simulate the liposome of natural erythrocyte membrane, which overcame the shortcomings of other blood substitutes, and it was a promising oxygen carrier.In this study, porcine hemoglobin was encapsulated with liposome and retention period was detected. Then it was transfused into dogs and cats as blood substitutes in animal clinical trials. The main contents was consist of three sections.(1)The reparation of porcine hemoglobin encapsulated with liposome was improved. Soybean lecithin and cholesterol were dissolved in ether solution, and then the hemoglobin solution was added. The mixed solution was placed into the rotary evaporator. It was rotated in the condition of that the temperature of water bath is25℃,the rotational speed is90rpm,the pressure is-0.02MPa.The ether was removed by slowly evaporating.The hemoglobin liposome was formed by ultrasonic bathing.The results show that:soybean lecithin and cholesterol mass ratio=2:1, oil phase and water phase volume ratio=2:1, the embedding rate is highest, reaching about37.65%.At4℃, procine hemoglobin liposome could preserved in a short period of time, but freeze-thaw caused the fractured of liposome, and porcine hemoglobin was disclosured. The porcine hemoglobin lipid would be degenerated at37℃.(2)When it was transfused into healthy dogs and cats, there was no hemolytic transfusion reactions in cats and dogs. The heart rate, body temperature, the number of erythrocyte was normal, it showed that it was safe and available to cats. However, the indicators of the number of white blood cells, urine protein, bilirubin, urobilinogen were increased. It indicated that liposome have some impact on the body’s immune system, kidneys, liver. (3)Compared with control group, the porcine hemoglobin injected the hemorrhagic dogs had a certain improvement on the function of the oxygen supply to the body in the loss of blood group. Compared with the pre-transfusion, some urine indicators were increased.This research was mainly to improve the preparation of liposome, the preservation of liposome,and the safety testing in dogs and cats were carried out,and oxygen function of liposome was detected. This study would lay a foundation for future research.
