Objective: Diabetes is a global public health problem,and its pathogenesis is affected by environmental,genetic and other factors.Studies have shown that the onset of diabetes is not only related to genetic polymorphisms of frequently stained genes,but also to genetic polymorphisms of mitochondrial DNA.There are ethnic differences in the genetic susceptibility of diabetes.Previous studies found that the incidence of diabetes in the Liannan Yao population is higher than that of the local Han population,and the typing of 15 autosomal STRs and 19Y-STRs in the Liannan Yao population proved that Liannan Yao population has a unique subpopulation inheritance Structure,but the relationship between T2 DM susceptibility gene TCF7L2 polymorphism and T2 DM in Liannan Yao population and pre-diabetes could not be confirmed during further investigation.This study analyzed the genetic information of the 60 genomic loci of mitochondrial DNA of Liannan Yao people and the polymorphism of related haplogroups,and explored the genetic relationship between Liannan Yao people and other ethnic groups.The establishment of the system provides theoretical support.Methods: The Expressmarker mtDNA-SNP 60 four-color fluorescence kit was used to multiplex the 60 mitochondrial DNA loci of 209 unrelated relatives of the Liannan Yao ethnic group,and PCR amplification was performed using the Gene Amp PCR System 9700.Calculate the frequency and frequency of alleles by direct counting method,use HaploGrep 2.0 platform to divide the haplotype and haplogroup of mtDNA of Liannan Yao people,and calculate the population genetic parameters(random match probability,Individual recognition ability,variability and coupling probability,etc.).According to the published data,the diversity of mt DNA haplogroups in the Liannan Yao nationality and12 other comparison groups were compared and analyzed.The genetic distance between populations was calculated using PHYLIP software,phylogenetic tree analysis was drawn using Poptree 2 software,and Past4 software was used for principal component analysis.Results:In this study,we obtained the locus information of 60 loci of mitochondrial DNA and the polymorphism information of related haplogroups in Liannan Yao population.A total of 46 mutation sites were detected in 209 Liannan Yao people,and a total of 1728 mutation sites were observed,including base mismatches,including 46 base conversion sites,and a total of 1612 observation sites;base transversion There are 4sites and a total of 118 observations.At the nt9824 site,simultaneous single nucleotide transitions and transversions(A / T / C)were observed,while at nt1719,nt2706,nt3348,nt4491,nt7600,nt9123,nt9477,nt11251,nt15784 sites,no more were detected Morphology.A total of 38 haplotypes were found in 209 Liannan Yao individuals.From the point of view of genetic differentiation coefficients of mitochondrial DNA haplogroups,the Fst genetic distance,phylogenetic tree and principal component analysis results all show that the closeness of the Liannan Yao nationality and the She nationality in Fujian is the closest,and it is also compared with the Yunnan minority,Hunan Miao and other southern minorities It is close,but it is far away from the ethnic groups in the northwest of Lhasa Tibetan and Kazakh in Xinjiang.Conclusion:(1)The mtDNA haplogroup of the Liannan Yao ethnic group has obvious genetic characteristics of the southern Chinese population,and is closer to the genetic distance of the southern ethnic groups such as Fujian She,Yunnan Yao,Hunan Miao,etc.The study of communication history provides a reference.(2)The mtDNA gene haplogroup polymorphism is of great significance in type 2 diabetes screening and prevention strategies.This study provides further screening and early prevention of type 2 diabetes mellitus susceptible mtDNA sites in Nanyao research Foundation.
