This paper applies RAPD (Random Amplified Polymorphic DNA) and SSR (Simply Sequence Repeats) assay to detect the genetic polymorphism of rat-like hamster {Cricetulus trition) populations (1984-1988) in Raoyang Hebei Province, in order to determine the relationship between the genetic variation and population dynamics The conclusions are listed as follows:1. This paper reports a new method to extract genomic DNA from the long preserved liver specimen of rat like hamster {Cricetulus trition), which fixed with formalin.The key to this method was the pretreatment of the specimen. The result showed that the purified DNA could be used in PCR using random primer, the amplification of microsatellite site.2. The selected primers were used to screen the rat-like hamster populations. According to the results of RAPD, the proportion of polymorphic loci, Shannon's index and Nei's index were analyzed to describe the genetic variation of rat-like hamster populations. It showed there was seasonal difference in genetic diversity of rat-like hamster populations and the genetic variation tended to be higher for the spring population, and lower for the autumn population in the same year. It should be noted that there existed positive correlation between genetic diversity and population abundance in different years.3. Based on the four pairs PCR primers of Cricetulus trition, and two pairs PCR primers which were designed by means of the sequence of Cricetus cricetus, the primers-AJ532562 were selected.
