The research of this subject mainly includes two parts,which are the multi-phase taxonomic study of two new microbes isolated from the Mohe frozen soil area and the Chishui River.The taxonomy,phenotype and chemical analysis of the strains are used to determine its taxonomy.status.A Gram-stain-negative,non-motile,nonsporulating,rod-shaped,orange-pigmented bacterium,designated strain FQM01T,was isolated from a subterranean sediment sample in Mohe permafrost area,China.Strain FQM01 T grew optimally at 25?,pH 7.0 and NaCl concentration of 0-0.5%(w/v).Analysis of the 16S rRNA sequence revealed that the strain FQM01T belongs to the genus Sphingomonas.The closest phylogenetic relative was Sphingomonas spermidinifaciens GDMCC 1.657T(97.6%),followed by Sphingomonas mucosissima DSM 17494T(97.2%).The DNA G+C content of the isolate was 66.9 mol%(HPLC).Strain FQM01T contained Q-10 as the predominant ubiquinone,and C18:1 ?6c and/or C18:1 ?7c,C16:1 ?6c and/or C16:1?7c,C16:0 and C14:0 2-OH as the major fatty acids.Major polar lipids were phosphatidylglycerol,diphosphatidylglycerol,phosphatidylethanolamine,phosphatidylcholine,sphingoglycolipid,and unidentified glycolipid.Only sym-homospermidine was detected as the polyamine.According to genotype,physiological and biochemical and chemical analysis,strain FQM01T is considered to represent a novel species of for which the name Sphingomonas floccifaciens sp.nov.is proposed.The type strain is FQM01T(=CGMCC 1.15797T=KCTC 52630T).A Gram-reaction-negative,colorless-pigmented,rod-shaped,aerobic,non-motile bacterium,designated GSA243 T,was isolated from water samples collected from Chishui River flowing through Maotai town,Guizhou,south-west China.Analysis of the 16S rRNA sequence revealed that the strain GSA243T belongs to the genus Rhodoferax and was most closely related to members of the species Rhodoferax saidenbachensis,Curvibacter fontanus and Polaromonas jejuensis with 96.64-98.35%sequence similarities.The major fatty acids are C16:0 and C16:1 ?6c and/or C16:1 ?7c(summed feature 3).The major respiratory quinone is ubiquinone Q-8 and the major polar lipid was phosphatidylethanolamine.The DNA G+C content is 62.2 mol%.According to genotype,physiological and biochemical and chemical analysis,strain GSA243T is considered to represent a novel species of the genus Rhodoferax within the family Comamonadaceae,for which the name Rhodoferax bucti sp.nov.is proposed.The type strain is GSA243 T(=CGMCC 1.16287T=KCTC 62564T).
