This study includes two major parts, one is the polyphasic taxonomy study of Qipengyuania sediminis gen. nov., sp. nov., which was isolated from Qiangtang Basin of Qinghai-Tibetan plateau, and the other is the polyphasic taxonomy study of Flavobacterium buctense sp. nov., which was isolated from Chishui River in Maotai Town. Through the analysises of genetic, phenotypic and chemotaxonomic data, the microbial taxonomic status of the two strains are determined.Strain M1T, which is belonging to the family Erythrobacteraceae, class Alphaproteobacteria, is named Qipengyuania sediminis according to the location where it was isolated. Cells are Gram-reaction-negative, facultatively aerobic, chemoheterotrophic, oval or short rod-shaped, oxidase-negative and catalase-positive. Growth occurs between 10 and 37? (optimum 30?), at pH 6.5-9.5 (optimum pH 8.0) and in the presence of 0-0.5% NaCl (optimum 0%). The dominant respiratory quinone is Q-10, phosphatidylcholine, phosphatidylethanolamine, phosphatidylglycerol, diphosphatidylglycerol, sphingoglycolipid, three unidentified glycolipids, one unidentified phosphoglycolipid and one unidentified lipid are the major polar lipids, C17:1?6c (44.2%) and C18:1?7c (13.7%) are the major fatty acids,the DNA G+C content of strain M1T is 73.7 mol%.The type strain, M1T(=CGMCC 1.12928T=JCM 301821), was isolated from a borehole sediment sample collected from Qiangtang Basin in Qinghai-Tibetan Plateau, China.Strain T7~T, which is belonging to the genus Flavobacterium, family Flavobacteriaceae, is named Flavobacterium buctense according to the location where it was identified. Cells are rod shaped, non-gliding motile, aerobic, Colonies are dark yellow, round and smooth, cells do not produce flexirubin-type pigments, oxidase and catalase tests are positive. Growth occurs between 15 and 30? (optimum 25?), at pH 6.5-9.5 (optimum pH 7.5) and in the presence of 0-0.5% NaCl (optimum 0%). Chemotaxonomic analysis reveals menaquinone-6 as the dominant respiratory quinone, polar lipids of strain T7~T includes phosphatidylethanolamine, four unidentified polar lipids, one unidentified phospholipid and one unidentified aminolipid and C]5:0(27.7%), iso-C15:0 (23.5%) and iso-C15:1 (11.5%)are the major fatty acids, the DNA G+C content of strain T7~T is determined to be 38.2 mol%.The type strain, T7~T (=JCM 30750=CGMCC 1.15216), was isolated from a freshwater sample collected from Chishui River in Guizhou Province, China.
