The research focus on the microbial polyphasic taxonomy study of two strains CHY01 and GSA152 isolated from fresh water of Chishui River,Maotai Town,Renhuai City,Guizhou Province.Through morphological characteristics such as morphology,physiology,biochemistry,chemistry etc.and genotype characteristics such as genome size,GC content,genome composition and so on to establish its taxonomy status.CHY01T is a gram-stain-positive,strictly aerobic,non-motile,endospore-producing strain.Strain CHYO1T grows optimally at pH 8.0 and 28?.The sequence analysis of 16S rRNA gene indicates strain CHY01T belongs to the genus Brevibacillus,the closest bacterial strain is Brevibacillus panacihumi,whose similarity of 16S rRNA gene is 97.81%.The major respiratory quinone was MK-7 and the major polar lipid was phosphatidylethanolamine and phosphatidylglycerol.The major fatty acids were iso-C15:0,C16:0,anteiso-C15:0,C15:1iso H/C13:0 3OH and C16:1?7c/C16:1?6c.Genome sequencing revealed a genome size of 5.74 M bp and a G+C content of 50.55 mol%.Pairwise determined whole genome average nucleotide identity(gANI)values and digital DNA-DNA hybridization(dDDH)values suggested that strain CHYO1T represents a new species,for which we propose the name Brevibacillus dissolubilis sp.nov.with the type strain CHY01T(=CGMCC 1.15916T=KCTC 33863T).GSA152T is a gram-reaction-negative,non-flagellate,aerobic,peach-brown-pigmented bacterial strain.Strain GSA152T was found to grow optimally at pH 7.0 and 25?.Phylogenetic analysis based on 16S rRNA gene sequences showed that the isolate belonged to the genus Rhodoferax.The closest phylogenetic relative was Rhodoferax saidenbachensis(98.42%).The major respiratory quinone was ubiquinone Q-8 and the major polar lipid was phosphatidylethanolamine.The major fatty acids were C16:0 and C16:1?6c and/or C16:1?7c.Genome sequencing revealed a genome size of 3.67 Mbp and a G+C content of 61.17 mol%.Pairwise determined whole genome average nucleotide identity(gANI)values and digital DNA-DNA hybridization(dDDH)values suggested that strain GSA152T represents a new species,for which we propose the name Rhodoferax bucti sp.nov.with the type strain GSA152T(=CGMCC 1.16288T=KCTC 62564 T).
