Pu’er tea is mainly planted by cutting breeding,and the survival rate of cutting tea seedlings without spraying is only about 15%.Plant rhizosphere microorganisms have a significant impact on plant growth.The use of synthetic flora instead of natural flora may improve the survival rate of cutting tea seedlings.The soil samples of one-year and two-year cutting tea seedlings in Pu’er City,Yunnan Province were collected.The physical and chemical properties were tested.After the bacteria in the soil were cultured,the culture results were amplified and sequenced.A total of 211 ASVS were obtained from the soil of one-year tea seedlings and 225 ASVS were obtained from the soil of two-year tea seedlings.A total of 36 strains were selected as the core flora,and other bacteria were divided into groups A and B through the species correlation network diagram to synthesize the artificial flora.The tea seedlings were cutted and the artificial bacterial suspension was used for root irrigation.In the 35 day growth of tea seedlings,the most serious defoliation occurred in the untreated soil group and the sterile soil group;The core flora+a group germinated first with the least defoliation and the highest survival rate of tea seedlings.The survival rate of tea seedlings in the core+a strain soil group is 4times that of the normal situation,which provides a certain scientific basis and technical support for improving the survival rate of Pu’er tea seedlings.Strain WGZ8~Twas isolated from the above Pu’er tea garden soil samples.The strain was rod-shaped,gram-negative,non motile and facultative anaerobic.The best Na Cl concentration is 0-1.0%,the best p H is 7.0,and the best temperature is 33°C.The only respiratory quinone is ubiquinone 10(Q-10).The genome size was 5.17 MB and the DNA g+c content was 61 mol%.According to the results of many taxonomic studies,WGZ8~Tbelongs to the genus Microvirga and is a new species of the genus.It is named Microvirga puerhi sp.Nov.The standard strain is WGZ8~T(=CGMCC 1.19171~T=JCM 35317~T). |
