Purpose:Experiments in Huayu Tongbian Decoction based on the research of the evaluation of clinical curative effect observation,to explore slow transit constipation(STC)method of the preparation of rat model,confirmed "activating blood circulation and removing blood stasis,detoxication and dredging collaterals " prescription drugs hold in STC rat colon the dynamics of change,studied the effects of Chinese herbs for activating blood circulation and removing blood stasis,detoxication and dredging collaterals on intestinal motility and expression of P2X2 receptor in colon tissue of rats with slow transit constipation,explored the possible mechanism of activating blood circulation and removing blood stasis,detoxication and dredging collaterals in the treatment of slow transit constipation.From molecules,immune and other multi-layer reveal "long illness blood stasis,blood stasis toxin impairing collaterals" is the pathogenesis on STC,reveal " activating blood circulation and removing blood stasis,detoxication and dredging collaterals " treatment and the mechanism of treatment on STC new utility targets.Material and method:1.Preparation of rat model of slow transit constipation2.The evaluation of STC after successful modeling,each model group were given Huayu Tongbian Decoction and Mosapride treatment intervention,model control group and the blank control group will not be any drug but the same volume of distilled water.3.The rate of carbon propelling determination.4.Using Western-blot and RT-PCR to detect the expression of P2X2 receptor in the intestinal mucosa and submucosa of distal colon tissue.5.Performing Statistical analysis by using SPSS17.0 statistical software.Results:1.Preparation of rat model of slow transit constipation The whole experiment for three rounds of circulation,a total of 91 days.Compared with blank control group,body quality growth of model group rats were less(P < 0.05), rat feces was cracked and short.First black defecation time of the model group rats were longer.Feces in the model group were lighter in weight for 24 hours(P<0.05).2.Effect of Huayu Tongbian Decoction on expression of P2X2 receptor in colonic tissue of rats with slow transit constipation2.1.The m RNA expression level of P2X2 in the model group and the control group.Compared with the blank control group,the m RNA expression of P2X2 in the model group increased significantly(P<0.01),each drug administration group compared with the model group,P2X2 m RNA varying degrees of lower(P < 0.01),compared with the western medicine control group,the m RNA expression level of P2X2 in the model group of Huayu Tongbian Decoction with middle dose and high dose were no difference(P>0.05),the low dose was not obvious(P < 0.05).2.2.The protein expression level of P2X2 in the control group and the model group.Compared with the blank control group,the relative gray scale values expression of P2X2 in the model group increased significantly(P<0.01),each drug administration group compared with the model group,the relative gray scale values of P2X2 varying degrees of lower(P < 0.01),compared with the western medicine control group,the relative gray scale values expression level of P2X2 in the model group of Huayu Tongbian Decoction with middle dose and high dose were no difference(P>0.05),the low dose was different(P < 0.05).Conclusion:1.The rat model of slow transit constipation was successfully prepared by rhubarb powder.The rat model of slow transit constipation was prepared by rhubarb powder water decoction,which was in accordance with the process of disease formation,can be successfully replicated for further animal experiments.2.The occurrence of constipation in rats with slow transit constipation may be related to the expression of P2X2 receptor in the distal colon.The occurrence of constipation in rats with slow transit constipation may be related to the increased expression of P2X2 receptor in the distal colon.3.Effect of Huayu Tongbian Decoction on the expression of P2X2 in distal colon of rats with slow transit constipation The mechanism of Huayu Tongbian Decoction on the treatment of STC in slow transit constipation may be related to the down-regulation of the expression of P2X2 receptor in rat distal colon.
