| Purpose:To evaluate the clinical efficacy of Huayu Tongbian decoction in combination with acupuncture for slow transit constipation.To observe Huayu Tongbian decoction combined acupuncture on Tianshu,Zhongwan,Shangjuxu,Zhigou,Geshu,Xuehai,Hegu therapy intervention effect of slow transmit constipation mice model,to explore the therapy of ghrelin,NO system,5-HT regulation function of the system.Material and method:Thesis One: Clinical Research on slow transmit constipation of Blood-stasis Syndrome by Huayu Tongbian decoction combined acupuncture68 patients were randomly divided into treatment group(34 cases)and control group(34 cases)according to the lottery method.The treatment group were treated with Huayu Tongbian decoction and acupuncture,the control group were given Mosapride Citrate Tablets and the acupuncture,2 groups were treated for 4 weeks and to observe its clinic curative effect.Thesis Two: The experimental research on the mechanism of Huayu Tongbian Decoction combined acupuncture in treating slow transmit constipation75 mice adaptive feed after 1 week,using the random number method is divided into five groups: blank control group,model control group,traditional Chinese medicine group,acupuncture group,combination of traditional Chinese medicine acupuncture group,each group of 15 only.In addition to the blank control group,the rest of the four groups of mice with compound diphenoxylate tablets,20 mg/kg dose to fill the stomach through the mouth,for four consecutive weeks of copy slow both transmissive constipation model mice.Then the Chinese medicine group was given Huayu Tongbian decoction to fill the stomach through the mouth.Acupuncture group was given acupuncture on Tianshu,Zhongwan,Shangjuxu,Zhigou,Geshu,Xuehai,Hegu intervention.Chinese medicine acupuncture group was given Huayu Tongbian decoction to fill the stomach through the mouth,at the same time acupuncture Tianshu,Zhongwan,Shangjuxu,Zhigou,Geshu,Xuehai,Hegu intervention intervention.To fill the stomach through the mouth saline model control group,blank control group without making processing.The intervention time,daily for two weeks.At the end of the time after the intervention,by examining each mouse small intestine propulsion rate at the end of the evaluation factors of carbon intervention effect of slow both transmissive constipation mice.By using immunohistochemistry and western blot method to detect CGRP,SP,VIP,e NOS,SOM,5-HT3 R,5-HT4 R and SERT expression level in colonic tissue in mice and the improvement of hemorheology test indicators.Results:1.Clinical Research on slow transmit constipation of Blood-stasis Syndrome by Huayu Tongbian decoction combined acupunctureAfter 4 weeks of treatment,the treatment group total effective rate was 94.12%;the control group total effective rate 82.35%,.Two groups total effective rate and cure rate were statistically significant(P<0.05).After treatment,the bristol stool form scale in both groups have significant improvement,with statistical significance(P<0.05).There was no significant difference of efficiency between treatment and control group(P>0.05).After treatment,the colonic transit test have significant improvement,with statistical significance(P<0.05).After treatment,the improvement of hemorheology test indicators in the treatment group was more significant than that in the control group,with statistical significance(P<0.05).The number of times of spontaneous defecation improved significantly in the treatment group after treatment compared with the control group,and the difference was statistically significant(P < 0.05).After treatment,the satisfaction score of the treatment group was significantly lower than that of the control group,and the difference was statistically significant(P < 0.05).2.The experimental research on the mechanism of Huayu Tongbian Decoction combined acupuncture in treating slow transmit constipation(1)Model evaluation: Compared with blank control group,the rest of the four groups of mice at the end of the carbon propulsion rate significantly decreased(p < 0.01),with statistical significance.(2)Groups of mouse small intestine propulsion rate at the end of the carbon results show that compared with blank control group,the rest of the four groups of mice small intestine propulsion rate at the end of the carbon decreased significantly(p < 0.01),there is statistical significance.Compared with model control group,traditional Chinese medicine(TCM)and/or acupuncture intervention group at the end of the mouse small intestine carbon propulsion rate increased significantly(p < 0.01),there is statistical significance;Compared with Chinese traditional medicine group,Chinese medicine plus acupuncture group at the end of the mouse small intestine carbon propulsion rate increased significantly(p < 0.01),there is statistical significance.Compared with acupuncture group,traditional Chinese medicine(TCM)plus acupuncture group at the end of the mouse small intestine carbon propulsion rate increased significantly(p < 0.05),with statistical significance.(3)Compared with blank control group,the rest of the four groups of mice expression of CGRP and VIP were significantly elevated in colon tissue(p < 0.01),there is statistical significance.Compared with model control group,traditional Chinese medicine(TCM)and/or acupuncture intervention group mice colon tissue expression of CGRP and VIP were significantly reduced(p < 0.01),there is statistical significance.Compared with Chinese traditional medicine group,Chinese medicine plus acupuncture group mice colon tissue of CGRP and VIP expression levels(p < 0.01),there is statistical significance.Compared with acupuncture group,traditional Chinese medicine(TCM)plus acupuncture group CGRP and VIP expression in the colon tissue levels in mice(p < 0.01),with statistical significance.(4)Compared with blank control group,the rest of the four groups of SP expression level was significantly lower in colon tissue of mice(p < 0.01 or 0.05),there is statistical significance.Compared with model control group,traditional Chinese medicine(TCM)and/or acupuncture intervention group SP expression level was significantly increased in colon tissue in mice(p < 0.01),there is statistical significance.Compared with Chinese traditional medicine group,Chinese medicine plus acupuncture group SP expression level increased in colon tissue of mice(p < 0.01),there is statistical significance.Compared with acupuncture group,traditional Chinese medicine(TCM)plus acupuncture group SP expression level increased in colon tissue of mice(p < 0.01),with statistical significance.(5)Compared with blank control group,the rest of the four groups of e NOS expression levels significantly elevated in colon tissue of mice(p < 0.01),there is statistical significance.Compared with model control group,traditional Chinese medicine(TCM)and/or acupuncture intervention group e NOS expression in colon tissue of mice were significantly reduced(p < 0.01),there is statistical significance.Compared with Chinese traditional medicine group,Chinese medicine plus acupuncture group mice colon tissue of e NOS expression levels(p < 0.01),there is statistical significance.Compared with acupuncture group,traditional Chinese medicine(TCM)plus acupuncture group mice colon tissue of e NOS expression levels(p < 0.01),with statistical significance.(6)Compared with blank control group,the rest of the four groups of mice colon tissue of SOM expression level decreased significantly(p < 0.01 or 0.05),there is statistical significance.Compared with model control group,traditional Chinese medicine(TCM)and/or acupuncture intervention group SOM expression levels significantly elevated in colon tissue of mice(p < 0.01),there is statistical significance.Compared with Chinese traditional medicine group,Chinese medicine plus acupuncture group SOM expression levels in the colon tissue in mice(p < 0.01),there is statistical significance.Compared with acupuncture group,traditional Chinese medicine(TCM)plus acupuncture group SOM expression levels in the colon tissue in mice(p < 0.01),with statistical significance.(7)Compared with blank control group,the rest of the four groups of mice colon tissue of 5-HT3 R,5-HT4 R expression level decreased significantly(p < 0.01 or 0.05),there is statistical significance.Compared with model control group,traditional Chinese medicine(TCM)and/or acupuncture intervention group 5-HT3 R in colon tissue in mice,significantly increased 5-HT4 R expression levels(p < 0.01),there is statistical significance.Compared with Chinese traditional medicine group,Chinese medicine and acupuncture group mice colon tissue of a five-HT3 R,5-HT4 R expression levels(p < 0.01),there is statistical significance.Compared with acupuncture group,Chinese medicine and acupuncture group mice colon tissue of a five-HT3 R,5-HT4 R expression levels(p < 0.01),with statistical significance.(8)Compared with blank control group,the rest of the four groups of SERT expression levels significantly elevated in colon tissue of mice(p < 0.01),there is statistical significanc.Compared with model control group,traditional Chinese medicine(TCM)and/or acupuncture intervention group SERT expression in colon tissue of mice were significantly reduced(p < 0.01),there is statistical significance.Compared with Chinese traditional medicine group,immunohistochemical method to detect the traditional Chinese medicine(TCM)+ acupuncture group SERT in colonic tissue expression levels in mice(p < 0.05),with statistical significance.Conclusion:1.Huayu Tongbian Decoction in combination with acupuncture to cure slow transit constipation have a better effect than treated with Maren Soft Capsule orally and the acupuncture.2.Huayu Tongbian decoction combined acupuncture on Tianshu,Zhongwan,Shangjuxu,Zhigou,Geshu,Xuehai,Hegu therapy intervention on slow transmit constipation mice has good effect on promoting defecation.3.Huayu Tongbian decoction combined acupuncture on Tianshu,Zhongwan,Shangjuxu,Zhigou,Geshu,Xuehai,Hegu therapy intervention on slow transmit constipation mice intervention mechanism may be related to regulating the colon tissue CGRP and VIP,SP,expression levels are closely related.4.Huayu Tongbian decoction combined acupuncture on Tianshu,Zhongwan,Shangjuxu,Zhigou,Geshu,Xuehai,Hegu therapy intervention on slow transmit constipation mice colon tissue e NOS can inhibit expression level,and the expression level of SOM has a promoting effect,the mechanism of action of the treatment shows slow constipation may be through regulating the SOM,indirect inhibition of e NOS expression level.5.Huayu Tongbian decoction combined acupuncture on Tianshu,Zhongwan,Shangjuxu,Zhigou,Geshu,Xuehai,Hegu therapy intervention on slow transmit constipation mice laxative effect may be through the colon tissue of 5-HT3 R,5-HT4 R,increase the combination of both and 5-HT play a role of enhance bowel movements,and cut in SERT expression,reduce the intake of 5-HT SERT,may be keep the colon tissue high 5-HT level is the importan role. |
